| Food and drug safety related to the vital interests of the masses. Due to the special purpose and higher value-added of the erectile dysfunction (ED) drugs, it has become the target of lawless elements to counterfeit and sele fake goods. Driven by interests, illegal traders add the ED drug in herbal sexual health products and pose health threats to consumers. Herbal sexual health products are complicated, and the added ED drugs are diversity and unknown. To solve this problem molecular imprinting technology, nanotechnology, self-assembly techniques and electrochemical techniques were integrated used for the first time preparing of the ED drug surface molecularly imprinted magnetic beads and layer-by-layer self-assembled molecular imprinted electrochemical sensor. The specific work is as follows:1. Research and application of highly selective tadalafil molecularly imprinted magnetic beads:Computer-aided design was used to compare three commonly used functional monomers and the final choice was 2-(trifluoromethyl) acrylic acid (TFMAA). The prepared MPs coated magnetic nanoparticles (MlPs-coated MNPs) showed high recognition ability and fast binding kinetics for tadalafil. Due to the magnetic properties of MNPs, the MlP-coated MNPs were able to achieve rapid separation by an external magnetic field. The resulting MlPs-coated MNPs were used as dispersive solid-phase extraction (DSPE) materials coupled with HPLC-UV for the selective extraction and detection of tadalafil from herbal sexual health products. The the recoveries were in the range of 87.36-90.93%with the RSD< 6.55%.2. Research and application of Sildenafil molecularly imprinted electrochemical sensor:The reduced oxide graphene (RGO) was used to modify glassy carbon electrode (GCE) by layer-by-layer self-assembly. We constructed conductive molecular imprinted membrane on the functional electrode surface using sildenafil as a template molecule, p-phenylenediamine as the functional monomer. The obtained highly sensitive and selective electrochemical sensor was successfully used to detect sildenafil in herbal sexual health products. The recoveries were in the range of 92.34.97.71%with the RSD<2.31%. |
