Xanthoceras sorbifolia Bunge is a kind of oil and medicinal plant that is peculiar to China, whose seeds are widely used in the development of bio-diesel. Discarded shells countain flavonoids, saponins, coumarin, sterol, fatty acid and other active physiological substances, research on the active ingredient is of great economic value and social benefits.Through the research on separation and purification techniques of flavonoids and saponins from Xanthoceras sorbifolia Bunge, it shows that: ethanol extraction is more effective than water extraction.The result of mass fraction of flavonoids is 2.15% and saponins 2.5%. Flavonoids purification should choose X-5 macroporous resin and AB-8 macroporous resin for saponins. Chromatography process parameters of the macroporous resin by the use of medium-high pressure preparative chromatography are that eluent: alcohol/water, velocity: 15 mL/min, detection wavelength:254,280,310 and 365 nm, gradient elution. The purity of the flavoids of the main peak is 90.5%, and the purity of saponins is 94.67%.The experimental study of anti-free radicals of Xanthoceras sorbifolia Bunge flavonoids and saponins indicates that saponins and flavonoids have a certain ability to resist free radical, of which the ability to resist hydroxyl radicals is obviously higher than that to resist ascorbic acid under the same concentrations. Saponins bacteriostatic experiment results show that the saponins have the inhibition effect on the staphylococcus aureus and Escherichia coli. IC50 is 0.494 and 0.366, respectively.Through the measurement of the oxidation rate that tyrosinase catalyzes L-dopa, we researched the inhitory effect that flavonoids and saponins have on tyrosinase activities in vitro. The results show that both flavonoids and saponins have inhibitory effect on tyrosinase. It is mixed inhibition, and michaelis constant is 1.05 mg/mL. The mass concentration of saponins is 0.36 mg/mL, inhibition rate reaches to 64.6%. It is noncompetitive inhibition, and michaelis constant is 1.25 mg/mL.The results of the test that flavonoids and saponins’ inhibition towards pig pancreatic lipase show that both can inhibit the activies of pancreatic lipase. While Xanthoceras sorbifolia Bunge flavonoids’ concentration is 0.27 mg/mL, the inhibition rate will be 75%. The maximum inhibition rate that the saponin has towards pancreatic lipase could be 87.5%. According to the researches of ultraviolet and fluorescence chromatographic study upon enzyme protein molecules, flavonoids and saponins from Xanthoceras sorbifolia Bunge can influence the activities of the enzyme by changing the enzyme conformation.By MTT method, and with flavonoids and saponins as materials, its inhibition effect on MGC-803 gastric cancer cell growth was determined. The results show that the flavonoids and saponins both have certain inhibition effect on MGC-803 gastric cancer cell. The inhibition enhances with the increase of drug concentration. While flavonoids concentration of Xanthoceras sorbifolia Bunge is 500μg/mL, the inbibition rate is 38.5%. While saponins concentration of Xanthoceras sorbifolia Bunge is 500μg/mL, the inbibition rate is 36.79%.
