Particulate matters pollution have been one of the hot topics in the area of environmental research in recent years, but there have been only a few studies focus on the microorganism contamination of particulate matters. Because the concentration of airborne microorganism is poor, it is very important to choose a suitable sampling method. Wastewater treatment plant is an important part of the city which may be polluted seriously by microorganism contamination in the surrounding air environment. So, the airborne microorganism sampling method and pollution characterization of airborne microorganism in a wastewater treatment plant were systematically study.This study firstly built a 40 L chamber and verified the best conditions of filter sampler is: 0.45 μm millipore filter, sampling flow is 10 L/min. The filter sampler combined with DAPI fluorescent count method can accurately detect the concentration of bioaerosol. And then it was based on the result of filter sampler detection of the concentration of bioaerosol to evaluated the sampling efficiency of TSP sampler in a 10m3 chamber. The experiment result indicated that the ratio of collected bioaerosol concentration by TSP sampler to collected bioaerosol concentration by filter sampler could reach to 0.7~0.84. It has shown that the sampling efficiency of TSP sampler is high. And TSP sampler could reach high sampling flow is more suitable for collect low concentration of bioaerosol. So TSP samples could be used to collect the bioaerosol.The analysis of High-throughput sequencing method of samples in Wastewater treatment plant has shown that the air of pretreatment period is mainly affected by influent, while the air of A2/O sewage pool is mainly affected by the A2/O sewage and the other parts of air may be affected by multiple factors comprehensive. On the level of phylum, the mainly composition of bacteria in air samples was Proteobacteria(49.33%)ã€Firmicutes(20.05%)〠Bacteroidetes(7.28%)〠Cyanobacteria(7.06%)〠Actinobacteria(4.39%) and Acidobacteria(1.94%), which was affected by the harsh environmental conditions in air. On the level of genus, Aeromonasã€Rhodospirillumã€Terrimonasã€Arcobacter were derived from the influent of wastewater treatment plant while Clostridiaceae and Sphingomonas were derived from the A2/O wastewater and sludge. Influent and A2/O sewage could be the source of the dominant pathogen such as Arcobacter butzleri, Escherichia coli and Serratia marcescens in air.The pollution characterization of ARGs in bioaerosol were also analyzed by PCR and RT-QPCR. The result shown that the level of airborne microorganism to carry ARGs was 102~104 times lower than sewage and sludge. The airborne microorganisms which carried sul2 genes were more likely to survive. This study has put forward a method to absolute quantitative the concentration of ARGs in air. |
