Targeted drugs and carrier have been a research hotspot in recent years. In this dissertation, liver targeting peptides SP94 (SFSIIHTPILPL) with relatively high targeting was selected in this study, and use gold nanorod as a transportation carrier, and then discuss some problems occurred in the process of synthesizing solid-phase synthesis and gold nanorod. On this basis, this paper builds a transport system of gold nanorod-targeting peptide, and studies the release of targeted features and platinum.The main research content includes:1. Taking crystal seed growth methods to synthesize gold nanorod, making NaBH4 as reducing agent, CTAB as surfactant seed solution. In the process of synthesizing gold bars, ascorbic acid was taken as reducing agent. This paper discusses some problems occurred by synthesizing gold nanorod, which need to pay attention to. Using reaction product to carry out UV-VIS and TEM tests, the result shows that the presence of HCl can obviously increase the productivity of gold nanorod, but some golden goal occurred. Both AgN03 and excessive crystal seed solution can cause incapability of inoculating seed growth.2. Taking solid-phase synthesis method to synthesize targeting peptide. This paper adopts the method of solid-phase synthesis, and takes DMF as solvent for synthesis, and then probes into some problems. After the detection of HPLC and mass spectrometry, the results show that the solid-phase synthesis can get polypeptides of relatively good purity (purity).3. Building gold nanorod-targeting peptide system. This paper adopts a new stability method of gold nanorod, which is different from traditional PEG. This paper uses structure of peptide segment of GCGC, and connects it to the surface of gold nanorod, and gets a stable gold nanorod-targeting peptide system. The test of cell targeting reveals that this system has excellent hepatoma carcinoma cell targeting, and the reaction of peptide-Pt(Ⅳ) and GMP shows that this system has excellent controllable release property of carrying platinum.
