| Conjugated linoleic acid has many kinds of biological activity,anti-cancer,anti atherosclerosis,weight loss,,regulating the body’s immune function, promote the growth and development and many other effects and highly concerned and favored by people, international sales showed a rising trend year by year。Conjugated linoleic acid at present mainly in the safflower oil as raw material to synthesize。But the raw material price is high,the low purity isomerization,isomer separation problems。 In this study,Hubei endemic plant Sapium sebiferum as raw material, linoleic acid content was 98% by means of separation and purification, and then conjugated linoleic acid content of more than 98% obtained by alkali isomerizationã€hydrolysis。 This plant Sapium sebiferum seeds contain a lot of unsaturated fatty acids, including alpha linolenic acid and linoleic acid content is the highest,respectively 39.30% and 30.77%。This study has been carried out on the early development of Chinese tallow acid alpha – linolenicã€linoleic acid is a by-product of its development,such as linoleic acid as raw materials from production of conjugated linoleic acid, can provide added value from product development,and new ways to find a low cost production of conjugated linoleic acid。This study first through the ester exchange reaction of fatty acid glyceride is transformed into fatty acid ethyl ester of stillingia oil,which lays the foundation for the further separation。By urea inclusion for separation of saturated and polyunsaturated fatty acids,and cuprous ion complexation extraction technology for the separation and purification of the mixture of polyunsaturated fatty acids from stillingia oil, ethyl linoleate content is more than 98%。The optimum technological conditions were determined by orthogonal experiments:the first urea inclusion mixed urea,urea inclusion of fatty acid and ethanol ratio(W/W/W) of 1.5:1:5.25,,the inclusion temperature is 0℃, the inclusion of 2.5hours. The two urea inclusion mixed urea,urea inclusion of fatty acid and ethanol ratio(W/W/W) of 4:1:16,the inclusion temperature-8 ℃,inclusion time is 7h。Under this condition,the content of linoleic acid and linolenic acid ethyl ester was increased from 65% to more than 98%, the yield is 35% or more, the use of complex sub copper ion extraction technology,the linolenic acid and linoleic acid were separated by linoleic acid and linolenic acid with different number of double bonds。The optimum technological conditions were determined by orthogonal experiments:t Cuprous chloride ionic liquid ratio(N/N) for 1:1.6, fatty acid ethyl ester and the complexing agent ratio(m/m) for 1:4.5,complexing temperature is 15℃, complexing time is 4 hours。Under this condition,the content of ethyl linoleate increased from 35% to more than 98%,the yield is 30%。 Finally, the conjugated linoleic acid was prepared by high purity linoleic acid as raw material。The effect of the solvent dosage,dosage of alkali and ethyl linoleate ratio and the effect of the inclusion temperature and time on the content of the product and the inclusion of conjugated linoleic acid。Through orthogonal experiments, the optimum reaction conditions were determined as follows: the ratio of KOH, linoleic acid ethyl ester and PEG-400(W/W/W) 1:2.5:6.25,,reaction temperature 120℃,reaction time was 3 hours, under this condition,the content of CLA reached more than 95%,the conversion rate reached 80%。 Established by GC,GC-MS,NMR of the fatty acid composition of stillingia oil,method for determination of linoleic acid,and conjugated linoleic acid structures and content。... |
