Beta agonists and beta blockers are a class of drugs which have the function of adrenaline synthetic phenylethanolamine, they were used in the treatment of bronchial asthma, cramps, hypertension and other symptoms in the clinic.The active groups stucture of these drugs can be combined with beta receptor of animal tissue cell membrane, then a series of physiological effect are occurred, the smooth muscle is expansion, lung capacity is increase, and the release of insulin will be interfered at the same time, leading to accelerate the decomposition of glycogen and fat.Animals are feeded this class of drugs in husbandry process, the feed conversion rate will improve, achieving nutrition redistribution, then improving the lean meat rate of animal, so this kind of drug was also known as the "clenbuterol". The residues of clenbuterol in food will cause serious harm to the human health, in the past 20 years, several countries and regions in the world have occurred incidents of clenbuterol poisoning, many countries had been prohibited to use clenbuterol in animal breeding process. The Chinese government had release policies of not allowed use of clenbuterol in animal breeding process since 1997, and formed corresponding monitoring management mechanism, in 2011 the Chinese government announced special clenbuterol rectification scheme, in order to strengthening monitor clenbuterol in the foodstuff, but there are still lacking of the detection method of new drugs or other similar drugs. So it is very urgent that build high efficiency detection technology based on a class of chemical characteristics, and it is wide application range.In this paper, by studying chemistry characteristics of the beta agonists and blocking, established a liquid chromatography tandem mass spectrometry technology for determination residue of 30 kinds of beta agonists and blocker in pig urine, liver, pork.Beta agonists and blocker drugs are a medium to strong polar compounds, their structure contains-NH group, they easily get the protons to hydrogenation peak in the ESI ion mode, in product ion scan, [M+H]+ will remove H2 O, and at the side of NH alkanes will fracture, benzene ring structure will lost halogen elements or hydroxyl structure, forming rich characteristics of the ions, among them [M+H-H2O]+ has the highest abundance. The 30 kinds of beta agonists and blocker drugs have wide polar span, and their ionization constants in strong alkaline conditions are close, with ultra high pressure liquid chromatography and the end sealing of C18 column, under the slowly lift elution strength, the 30 kinds of compounds are obtained fast and efficient separation. Test shows that the enzymolysis effect of beta agonists and blocker drugs is fairly between the glucosidase enzymatic hydrolysis under 55 ℃ four hours and 37 ℃ sixteen hours. After comparing the different solid phase extraction column and extraction conditions, MCX solid phase extraction column is suitable for 30 kinds of analyte purification and enrichment in pig urine, pork liver, pork.In this paper established rapid determination method of 30 kinds of beta agonists and blockers drugs residue, quantitative limit is 0.1 ~ 1.0 μg/kg in the three kinds of matrix, the recovery is 60.0% ~ 120.0%, relative standard deviation is 0.8% ~ 19.6%. Analysis time of the method is short, the method has been reliable of qualitative and quantitative, and it can be used determination of 30 kinds of beta agonists and blockers drugs residues in pig urine, pork liver, pork.
