Okara contain lots of protein and fat, which are the greatest by-product in industry of processing to-fu, soymilk,etc. However, it has not been paid great attention because of the low calorie and the poor taste, which has caused a serious waste of resources. The paper is studied on using reverse micelles method to extract proteins and oil from okara, which is not only simple process, high protein extraction rate, but also avoids the problem of protein denaturation.According to the properties and characteristics of okara proteins, the paper selects the DTAC reverse micelle system, which has the characteristics of non-toxic,rapid preparation and high extraction efficiency of okara proteins.The influence law of forward extraction ratio of okara protein by the addition amount of okara, DTAC concentration, extraction time, KCl concentration, Wo, buffer pH and extraction temperature was researched, obtained by orthogonal experiments, the optimum extraction conditions of okara protein, which okara dosage of 0.025 g/mL, Wo value of 35, DTAC concentration of 0.09 mol/L, extraction time of 30 min, pH value of 7.5, KCl concentration of 0.15 mol/L, the extraction temperature is 35℃. The verification experiment show that the average forward ratio of okara protein is 89.65%.And the influence law of after extraction ratio of okara protein by buffer pH, KCl concentration, oscillation time and extraction temperature were also researched, obtained by the response surface design of experiment, the optimum conditions on after-extraction conditions of okara in reverse micelles were when the pH of the aqueous phase is 6.50, the KCl concentration is 1.95 mol/L and the reaction time is 1.63 h, the after-extraction rate in theory of okara proteins is 87.68%, the real after-extraction rate is 86.04%.The results show that the response surface design model is remarkable and in well fit.
