Rhubarb is the root of Polygonum Rheum palmatum L.、Rheum tanguticum Maxim.ex Balf.or Rheum officinale Baill. The optimal process conditions of extraction for sennoside A from Rhubarb were researched in this article, as well as the quality standard. The content and resμLts of the experiments are presented in detail.1. Establishment of the assaying method for sennoside AObjective To establish assaying method for sennoside A which used for treating diarrhea in Rhubarb. Methods The primary functional ingrident of Rhubarb is measured by R-HPLC.The parameter as follows:chromatogram column is reverse AlltimaTMC18(5μm,250mm×4.6mm), LC20 pump, SPD-M20 A detector and measured wavelengh is 340 nm,solvent is methanol and 0.1% of phosphoric acid(45:55),velocity of flow is 1.0mL/min,column temperature is 30℃.In this measured condition,the time of apex of sennoside A is 18.477 min. ResμLts The concentration of sennoside A ranged from 0.01 to 0.05mg/mL had better linear relationship with the peak area. And the resμLts of precision, stability, reproducibility and recovery were consistent with the regμLations. Conclusions This method is able to determine the content of sennoside A in Rhubarb because it is convenient, fast, and accurate.2. Extraction process of the sennoside AObjective To obtain the optimal process conditions of extraction for sennoside A in Rheum tanguticum Maxim.ex Balf. Methods The factors influenced the extraction effect seriously were carried out first throμgh single factor experiment, including the concentration of NaHCO3 solution, the dosage of NaHCO3 solution and extraction time. The response surface experiments were done on the basis of the single factor resμLt, in which the content of sennoside A was taken as target. ResμLts The best optimal conditions were as follows: 0.25 g powder of Rhubarb was weighed, and dissolved in 40.8mL, 0.15% of NaHCO3 solution, μLtrasonic extraction 40.5min, filtered. The content of Sennoside A can reach to 0.38% while measured by HPLC.Conclusions The process was stable, higher extraction rate Sennoside A lay the foundation for the industrial production of Rheum Sennoside A’s.4. Quality Standard of RhubarbObjective To study the quality control methods of Rhubarb. Methods The gallic acid was identified throμgh TLC. The main ingredient of Rhubarb such as emodin rhubarb, rhubarb phenol, rhein, aloe-emodin content physcion arsenic and content of heavy metal such as arsenic, lead, mercury, cadmium were checked according to TLC, HPLC, atomic fluorescence analysis and graphite furnace analysis. ResμLts The TLC resμLt was observed in 365 nm, and the speckle was orange corresponding with the reference showed. The developer was ammonia fumigation and the speckle turned into red when placed under the observation of fluorescent.The total content of those main ingredient of Rhubarb was 1.64% respectivetly determined by HPLC, it has reached the requirement of the Pharmacopeia and the national quality standards. The content of heavy metal was in line with Pharmacopoeia requirements. Conclusions The quality can be controlled better throμgh the items including identification, detection, and assaying. It will be the foundation of more research about total tannins.
