Iris tectorum roots are the root of Iris of Freesia.Withing function of anti-oxidation and cracking to produce aroma, Flavonoids are the main active ingredient of the orris root.So,it has great prospects for research and exploitation.Firstly, this paper has studied the extraction and purification process of Iris root flavonoids. And the process conditions of the extraction and purification of Iris root flavonoids are optimized,successively. Then, by the method of modern chromatographic, spectroscopic and MS techniques,several Iris root flavonoids were purified and identified.The optimal method of separation and purification are obtained.The main ingredient of the orris root flavonoids are determined. Finally,the affection of flavonoid monomers and mixture obtained by different extraction solvent to cigarette quality were studied.(1) Extraction process of orris root flavonoids were optimized.The optimal process conditions founded were temperature 80℃,pH 7.0,ethanol concentration 70%,volume ratio of solvent to sample lOmL g-1,extraction of 2 times and 2 hours for each time.The results showed this method is optimal.The extraction rate was above 5% and the flavonoids content of extract was above 13%.(2) By using AB-8 macroporous resin,orris root flavonoids were separation and purification.Adsorption rate, desorption rate and recovery rate as index to investigate various factors’influence for separation and purification of orris root flavonoids with method of combining static adsorption and dynamic desorption.Result show the optimal condition were Sample solution pH 6.0,eluent concentration 70%,Sample solution volume 50mL,eluent current 2mL/min,eluent volume 140mL.Under the conditions,recovery rate of total flavonoids was 75.12% and the purity of total flavonoids was 54.2%.The optimal conditions are reasonable and feasible, and suitable for industrial production.(3)Six flavonoids monomer compounds were isolated from orris root,including three kinds of glycosides:tectoridin, Iristectorin B, IristectorinA;three kinds of aglycone compounds:tectorigenin,Iristectorigenin A, Iristectorigenin B.(4)The affection of each flavonoid monomers and different solvent extracts to cigarette quality were studied. Orris root flavonoids obtained by separation and purification has improved aroma and taste of cigarettes to some extent.Among of them,the extracts rich in flavonoid glycoside are especially effective.Flavoring effect of extract is better than monomer compounds taking into account the effect of smoking.It suggested orris root flavonoids complex better than a single ingredient in flavoring effect.So it is not the higher purity of the flavonoid monomer, the better flavoring effect.The study provides a optimal method to prepare orris root flavonoids active ingredients by extraction and separation. Simultaneously,this study provide guidance theory and methods of flavonoids orris root application in tobacco.
