Six new ruthenium-arene complexes with flexible ligands have been synthesized and characterized in this thesis.Six complexes [Ru(C10H14)Cl]2(C17H20N4)2Cl2(1),[Ru(C10H14)Cl]2(C17H20N4)2(CF3SO3)2(2),[Ru(C10H14)Cl]2(C18H22N4)2(CF3SO3)2(3),[Ru(C10H14)Cl]2(C15H18N6)2(CF3SO3)2(4),[Ru(C10H14)Cl]2(C16H20N6)2(CF3SO3)2(5) and [Ru(C10H14)Cl]3(C18H21N9)(CF3SO3)3(6) have been synthesized and recrystallized by stirring [RuCl2(p-cymene)]2and1,3-bis(1-imidazol-1-ylmethyl)-2,4,6-trimethylbenzene (m-bitmb),1,4-bis(1-imidazol-1-ylmethyl)-2,3,5,6-tetramethylbenzene (p-bitmb),1,3-bis(1-trazol-1-ylmethyl)-2,4,6-trimethylbenzene (m-bttmb),1,4-bis(1-trazol-1-ylmethyl)-2,3,5,6-tetramethylbenzene (p-bttmb),1,3,5-tris(trazol-1-2,4,6-trimethylbenzene)(tttmb), respectively.The crystal structures of complexes1and2have been characterized by single crystal X-ray diffraction analysis, and they have nearly the same components but different configurations.1is like a "boat" while2is like a "chair". There are Ï€-Ï€ interactions between the molecules of1, and there are no such interactions in2.For1-6, the hydrolysis and interactions with guanine have been characterized by UV-vis. For1and2,1H NMR is used to characterize the hydrolysis and the interaction with9-EtG. The strucutre of complexes make a key role in their hydrolysis and the inter actions with base group. It is easier to hydrolyze and interact with base group when the leaving group in open environment.Cyclic Voltammetry (CV) studies have been used to characterize the redox properties of complexes1-6, they exhibited reversible, quasi-reversible, or irreversible oxidation waves that are assigned to the Ruâ…¡â†'Ruâ…¢ oxidation. The structure and configuration of ligand can make affect to the redox potential. All complexes are more stable in Ruâ…¡ than Ruâ…¢, and there is no electron transfer between metal nuclear through ligands.The interactions of complexes1and2with ct-DNA were studied by circular dichroism spectroscopy. The secondary structure of ct-DNA has been retained after complexes1and2interacted with ct-DNA.The cell toxicity of complexes1-6to some tumor cells were studied by MTT assay and it was showed that some complexes were cytotoxic to some tumor cells.
