Lysophospholipid is efficient and stable emulsifier, can be used in food processing, feed, pharmaceuticals, cosmetics, and so on. In this study, ethanolysis of phosphatidylcholine(PC) catalyzed by phospholipase A1(PLA1) was operated in solvent and solvent-free system. The reaction factors affecting the PC ethanolysis and optimum conditions were studied. Furthermore, the influence of other alcohols on the PC ethanolysis catalyzed by PLA1 was discussed.Preparation of the lysophosphatidylcholine(LPC) through ethanolysis of PC by PLA1 in solvent system was studied. Effects of the reaction factors, including PLA1 dosage, water content, substrate ratio, reaction temperature and solvent content, on the LPC conversion of the PLA1-catalyzed ethanolysis of phosphatidylcholine were studied. Based on the single-factor experiments, the reaction conditions were optimized through response surface method. The optimal condition was as follows: PLA1 dosage 40 uL/g(phospholipase A1/PC, v/w), water content 25 uL/g(water/PC, v/w), substrate ratio 1:3(PC:ethanol, mol/mol), solvent content 1:2(PC:hexane, w/v), reaction temperature 30 ℃, reaction time 3.55 h. At these conditions, the content and convertion rate of LPC was 70.1% and 98.32% respectively.Five kinds of alcohols were added in ethanol respectively to study the influence on the ethanolysis of PC by PLA1 in solvent system. When n-propanol was added, the alcoholysise was inhibited by compared to enthanolysis which no other alcohol was added. When butanol was added, there was no influence on the alcoholysis. When ethanediol, 1,2-dihydroxypropane and glycerol was added respectively, the alcoholysis was promoted.Preparation of the LPC through ethanolysis of PC by PLA1 in free solvent system was studied. Effects of the reaction factors, including substrate concentration, PLA1 dosage, reaction temperature and water content, on the LPC conversion of the PLA1-catalyzed ethanolysis of PC were studied. Based on the single-factor experiments, the reaction condition was optimized through response surface method. The optimal condition was as follows: substrate concentration 3.5 g/mL(PC/ethanol solution, w/v), PLA1 dosage 54.6 uL/g(PLA1/PC, v/w), reaction temperature 30 ℃, water content 14.9 uL/g(H2O/PC, v/w), reaction time 7 h. At these conditions, the content and convertion rate of LPC was 60.5% and 97.2% respectively.Five kinds of alcohols were added in ethanol respectively to study the influence on the enthanolysis of PC by PLA1 in free-solvent system.
