The research for the interaction of small molecules with DNA are becoming one of hotspots in the field of molecular biochemistry since the discovery of cantineoplastic activity of cis-[Pt(NH3)2Cl2]. G-quadruplexes exist in some important parts of human genome including chromosome telomeres and some oncogene. In recent decades, the studies on anti-cancer drugs targeting G-quadruplex DNA are attracting great interest. Considerable evidence demonstrated that small molecules compounds could promote the formation of G-rich sequences and stabilize the G-quadruplex structures, which could inhibit the occurrence of tumor through the prohibition of replication of cancer gene or telomerase activity. The investigation of the interactions and mechanism between small molecules and DNA are important for exploring the application of complex in the field of cantineoplastic Pharmaceuticals, molecular biochemistry, bioengineering technology, etc. Transition metal complexes play an important role in nucleic acid chemistry as their diverse applications such as footprinting agents, sequence specific binding, structural probes, and therapeutic agents. The interaction between transition metal complexes and DNA, espectially transition metal complexes containing phenanthroline or shiff base ligands are more attractive.In this paper, the copper(II) complexes containing phenanthroline and shiff base ligand were synthesized, and the interaction with DNA were studied. This research includes three parts:In the first part, the discovery and development of nucleic, the basic knowledge of DNA structures and G-quadruplexes are elaborated. And then, the modes of interaction as well as common research methods between compounds and DNA are also illustrated. Finally, it presented research purposes and the main content of thesis.In the second part, three copper(II) complexes containing phenanthroline were synthesized and characterized by 1H NMR, MS, IR, elemental analysis and crystal structure analysis. Their photophysical and electrochemical properties, the interaction with ctDNA, the mechanism of DNA cleavage and the cytotocity of the complexes were studied.In the third part, based on the previous work, shiff base ligand and its copper(II) complex were synthesized and analyzed by crystal structure. The sensing of Cu2+ by shiff base liagand and the electrochemical properties of the complex were studied. And their photophysical properties, the interaction between compounds and DNA, the gel electrophoresis and cytotocity were also investigated.
