In the chapter one, first of all, the related technical background were introduced, Such as the preparation of gold nanoparticles and their application in biomedical, DNA markers, tumor cells, chemiluminescence, fluorescence and laser confocal microscopy, etc., understanding of the current research status and detection technology, these have a guiding significance for our research subject. Finally we introduced the basis of this topic research and content.In the chapter two, Gold nanoparticles (AuNRs) were prepared, they can relate with a variety of biological molecules when Gold nanoparticles as markers. Through some processing of DNA, we established the system of functional nucleic acid aptamer gold nanoparticles to detect ATP and it can combine with cells and further test the content of ATP in the cells. Detection limit of 4.0 × 10-8 M in the concentration of ATP, and it can detect 200-5000 Ramos cells.In the chapter three, an ATP electrochemiluminesce sensor was designed based on as-prepared cadmium sulfide (CdS) quantum dots, this chemiluminescence method has fast response to ATP. When ATP exists in the the system, it can combine with ATP aptamer chain, thus releasing the strand of CdS-DNA’, CdS can produce ECL signal in PBS containing 0.1 M KCl and 0.05 M K2S2O8. By the changes of ECL intensity, we can determine the concentration of ATP. After the optimal experimental conditions were optimized, this sensor has a response when the concentration of ATP rang from 2.0 × 10-9M to 1.0 × 10-7M, We can modify DNA on the piece of gold to measure the fluorescence, when the concentration of ATP rang from 2.0 × 10-9-1.0 × 10-7 M, this sensor has a response too, such we think this method has a good application prospect in the field of biological analysis.
