To research the scale-up thermostable xylanase production by Thermomyceslanuginosus DSM10635, a solid state fermentation(SSF) process was designed andoptimized by cultivation of corncob. T. lanuginosus DSM10635producesthermostable xylanase, which showed thermotolerance at75℃with half-life timeover30mins. A clever device was designed with intermittent ventilation with bettermoisture retention. Meanwhile, the culture media were optimized to be cheaper.In laboratory scale testing, the average activity and the highest activity ofxylanase obtained after7days cultivation of dry corncob were34831IU/g and53710IU/g, respectively. At that optimized culture condition, beef extract was removed over80%, and the culture media was120g/bottle compare with the60g/bottle in theoriginal SSF. Obviously, the yield of thermostable xylanase productivity was muchhigher than the yield under original culture conditions.In pilot scale-up testing, the nutrient solution of media were optimized to bemore unexpensive with50%-off yeast extract on the laboratory scale testing.Theaverage activity and the highest activity of xylanase obtained after7days cultivationof dry corncob were8237.4IU/g and13100IU/g, respectively. This is the first reportabout xylanase industrial production by SSF in China.
