In recent years, with the level of computer hardware continuous improving, micro manipulation system was improved. Micro operations location technology is one of the important features of micro operation system. In the process of micro operations, because the operation target is very small, precise positioning is very difficult, an accurate positioning technology is needed imperatively. In this paper, target tracking theory was studied and a method of micro image positioning and resolution strategies were put forward novelly.Firstly, micro visual system of micro manipulation system determines the accuracy of target localization, the micro objective lens is an important part of micro visual system, the optical properties of micro objective lens was studied. Optical transfer function of the micro objective lens was studied after the optical properties. The ten times micro objective lens of micro visual system was used in experimental process, optical transfer function with non-overlapping phenomenon was acquired, the micro target location technology is improved.Secondly, micro objects have the characteristic of less feature information, more noise and more vulnerable to external environment influence, in this paper, a micro object tracking algorithm based on optical transfer function is proposed. Optical transfer function of the micro objective lens enhanced frequency domain characteristic of micro image, reduced the noise of the image information, improved the quality of the image and enhanced the accuracy of micro image location.Thirdly, an experiment of the micro image processed by optical transfer function was carried out. Analyze the shortage of the Butterworth filter, calculate and analyze the signal-noise ratio of the micro image after different processing methods. The result proved that the noise of the image information was reduced, the quality of the image is improved after the processing by optical transfer function.In additionally, an experiment of micro target positioning by a micro target tracking algorithm based on optical transfer function was carried out. Analyze the traditional template matching algorithm, Kalman filter and target tracking algorithm based on watershed segment. Because the target tracking algorithm based on watershed segment has the characteristics of light resistant and deformation resistant, the same micro images were processed by the target tracking algorithm based on watershed segment. The result proved a micro target tracking algorithm based on optical transfer function was effective.
