Nifedipine(NIF),which was dihydropyidines calcium channel blocker, was widely used as antihypertension in clinic. It had antiangina and lowing blood pressure effects,had low side effects. In order to facilitate hypertension suffers and improve efficacy, we studied and prepared NIF sustained release liquid filled capsules.In the preformulation studies, physical and chemical properties of NIF such as equilibrium solubility, stability of aqueous solution and apparent oil-water partition coefficient were investigated. We initially studied release of NIF.The formula and technics of NIF sustained release liquid filled capsules was the focus of this article. Based on the property of the excipients,we used Gelucire 1 and hexadecanol as the sustained release excipients .To solve the problem that the release of NIF speeded up in place, HPMC was added into the formula. Orthogonal design was carried out to optimize the formulation. The studies identified the final formula of NIF sustained release liquid filled capsules.The release profile of enlarge test samples were good.The topics chosen artificial neural network algorithm BP algorithm optimization results show that BP network and orthogonal design methods, forecast and optimize its value closer to the experimental values. Choose the more efficient price formula to experiment, it saves a lot of time.By using DSC and X-ray diffraction analysis,NIF in formulation was amorphous state and formed solid dispersion. The stress testing showed that Solid dispersion of NIF were stable when exposed to high tempertutre and humidity.In order to control the quality, quality standard of NIF sustained release liquid filled capsules(including identification, related substance, content, dissolution) was developed.To evaluate the release profile of NIF sustained release liquid filled capsules in vitro, multimodal analysis was done.Peppas equation’s data was closest to reality.According to the n value of Peppas equation ,we can judge the release mechanism of NIF sustained release liquid filled capsules was synergy results of diffusion and framework corrosion.
