The purpose of the studyThrough the observation of Xiaoyao Powder (compound containing estrogen like medicine Angelica) effect on MCF-7cells induced by human breast cancer in nude mice, to study the dispersion effect on the treatment of breast cancer and the possible mechanism of xiaoyao.Research methods1. human breast cancer cell line MCF-7established human breast cancer in nude mice model of breast cancer. After the modeling, the36nude mice were randomly divided into6groups (single drug Angelica group, Xiaoyao powder group, high dose and low in Xiaoyao dispersed Angelica group and control group, with6rats in each group), respectively, according to18.6g/kg,12.0g/kg,24.0g/kg,48.0g/kg,39.1g/kg dosage and saline intervention, continued for28days. Measurement of tumor periodic long, short diameter, calculation of tumor volume, tumor growth curve; the twenty-ninth day, mouse eye blood, serum preparation, detection of serum estrogen, progesterone and ELISA assay; removal of tumor, the tumor weight, tumor inhibition rate was calculated; chemical method to detect the expression of estrogen receptor and progesterone of breast receptor in immune tissues.2. Kunming clean female weanling mice60, were randomly divided into6groups (single drug Angelica group,Xiaoyao powder group, high dose and low in Xiaoyao dispersed Angelica group and control group),10rats in each group, respectively with18.6g/kg,12.0g/kg,24.0g/kg,48.0g/kg,39.1g/kg and normal saline intervention, sustained drug delivery the14day,24h after the last administration, broken neck treated mice, immediately cut weighed uterus, uterus coefficient and calculation (uterus coefficient=uterine wet weight/body weight×100%). Result1. by MCF-7cell culture, breast cancer in nude mice model, the average tumor formation time was6days. Be different interventions, angelica group the tumor volume growth rate in the intervention of ninth days up to518.4±63.7mm3, far more than the control group482.3±58.2(P<0.05), to the end of the experiment, the volume reached2235±247.3mm3, while the control group with the same period in Xiaoyao Powder volume, high dose group of tumor is decreased, especially the high dose of Xiaoyao Powder group (P<0.05); Xiaoyao Powder in middle dose group, high dose group and Xiaoyao tumor weight was lower than control group, Xiaoyaosan tumor rate up to29.19%in high dose group, anti; in Experiment4weeks later, changed each animal estrogen, progesterone levels, compared with the control group, single drug Angelica group estrogen level is significantly higher than that of control group (P<0.05), and Xiaoyao Powder low, middle and high dose group, estrogen levels were decreased (P<0.05);detection of ER, PR receptor, Xiaoyao powder expression level in each dosage group in breast cancer tissues was significantly higher than that in the control group, the differences were statistically significant (P<0.05).2, single medicine Angelica group wet weight of uterus and uterine index compared with the control group (P<0.05),are large, and Xiaoyao Powder in high dose group, mouse uterine wet weight and their organ coefficients are smaller than that of the control group, compared with the normal saline group,(P<0.05).Conclusion1. Xiaoyao San can inhibit breast cancer growth in nude mice; the mechanism may be related to decrease the levels and activity of estrogen in the body, and the expression increased breast cancer in nude mice with estrogen receptor and progesterone receptor.2. Xiaoyao Powder without promoting mouse uterotrophic effect, does not have the effect of estrogen, angelica has plant estrogen like effects, but in Xiaoyao Powder does not exhibit the estrogen like effects of prescription.
