ObjectiveThis study aims mainly to:(a) observe the effect on the red blood cell (RBC), hemoglobin (Hb) and immature reticulocyte fraction (IRF) performances of the blood-deficiency modeled rats when treated with different twisting-twirling directions of twirling reinforcing and reducing manipulation;(b) explore further the relationship between the twisting-twirling and reinforcing-reducing manipulation of acupuncture and the twisting-twirling direction;(c) provide experimental evidence for standardization of the twirling reinforcing and reducing manipulation.Method80healthful male SPF Wistar rats with weight ranging from180g to200g, were randomly divided into7groups:group A (blank group), group B (model group), group C (acupuncture group), group D (right-hand-forward twisting group), group E (right-hand-backward twisting group), group F (left-hand-forward twisting group) and group G (left-hand-backward twisting group). We arrange10rats in the former two groups and12rats in the last5groups (treatment groups).The rats in group A are not treated, while the rats in the rest groups are established to be the blood-deficiency models using tail-bleeding and food restriction. After that, the rats in groups C, D, E, F and G were acupunctured with different methods, i.e.,(a) acupuncture with no twisting for group C;(b) right thumb twisting forward heavy for group D;(c) right thumb twisting backward heavy for group E;(d) left thumb twisting forward heavy for group F; and (e) left thumb twisting backward heavy for group G. At the same time, the rats in groups A and B were arrested with the same catch stimulation. The needling deep is about5mm, twirling magnitude is about360°/second, the frequency is about60beats/min, the maximum intensity is about0.08N, do1min×2times on each side, the needle remaining time is20min. Do treatment every other day with10times totally. Finally the RBC, Hb, IRF performances of each rat are measured using the XE-5000automated hematology analyzer at different time periods:(a) before modeling;(b) one day before the acupuncture treatment;(c) acupuncture treatment for7days;(d) acupuncture treatment for14days; and (e) acupuncture treatment for21days respectively.Result1The effect on the RBC performance with different treatmentsBefore modeling, the RBC value has no significant difference among all the rats in each group (p>0.05). At the day before the treatment, the RBC of all the groups, except the blank group (A group), decrease significantly (P<0.01), while the other groups have no significant difference (p>0.05). After7days treatment, the RBC of each group are measured and compared respectively, the comparison results are as following:(a) the group B was the lowest (P<0.05);(b)no statistical differences (P>0.05) were found among the groups Cã€D and F;(c) no statistical differences (P>0.05) were observed among the groups A. E and G;(d) The RBC of the first three groups were obviously higher than those of the latter three groups (P<0.05). After14days treatment, the RBC of all the groups, except the blank group (A group), increase significantly (P<0.01), while the other six groups have no significant difference. No statistical differences (P>0.05) are observed among all the groups after21days treatments (P>0.05).2The effect on the Hb performance with different treatmentsBefore modeling, the Hb value has no significant difference among all the rats in each group (p>0.05). At the day before the treatment, the Hb values of the six groups with blood-deficiency models decrease significantly compared with group A (P<0.01), but there is no significant difference among these six groups (P>0.05). After7days treatment, the rats in group B have an obviously lower Hb values than the other six groups (P<0.05). Meanwhile we can also get the following comparison results:(a) no statistical differences (P>0.05) were found among the groups Cã€D and F;(b) no statistical differences (P>0.05) were found among the groups Aã€E and G;(c) The Hb of the former three groups are obviously higher than those of the latter three groups (P<0.05). After14days treatment, the Hb of the first group are lower than the other six groups (P<0.05), but there is no statistical differences among these six groups (P>0.05). No statistical differences (P>0.05) are existed among all the groups after21days treatments (P>0.05).3The effect on the IRF performance with different treatmentsBefore modeling, the IRF value has no significant difference among all the rats in each group (p>0.05). At the day before the treatment, the IRF of all the groups, except the blank group (A group), increase significantly (P<0.01), while there is no significant difference among other six groups (p>0.05). After7days treatment, the IRF of all the groups, except the blank group (A group), decrease significantly (P<0.05), while there is no significant difference among these six modeling groups (p>0.05). No statistical differences (P>0.05) were observed among all the groups after14and21days treatments.Conclusion1Heavy-thumb forward twisting is "reinforcing method", while heavy-thumb backward twisting is "reducing method", no matter using the right or the left hand;2The status of the body plays a decisive role on the effect of the twisting-twirling and reinforcing-reducing manipulation. More specifically, in the case of blood-deficiency, any acupuncture treatment can produce "reinforcing" effect;3For the rat with blood-deficiency model caused by tail-bleeding and food restriction, acupuncture treatment can improve its peripheral blood circulation, but has no obvious effect on the bone marrow function. |
