Objective:Develop Type2diabetes mellitus (T2DM) rat model to investigate Liu Wei Di Huang Tang on fasting blood glucose in type2diabetic rats (FBG), fasting serum insulin (FINS), serum cyclic adenosine monophosphate (cAMP) levels and liver phosphatidylinositol-3-kinase catalytic subunit (PI3KP110), affect the level of phosphorylation of protein kinase B (Akt) and its protective effect on the islets, explore the pathogenesis of type2diabetes, and research from the molecular level Liu Wei Di Huang Tang intervention type2diabetes mechanism of action.Methods:High-sugar high-fat diet combined with intraperitoneal injection of45mg/kg streptozotocin (STZ) method to establish type2diabetic rat model, the establishment of the control group, model group, metformin group, Liu Wei Di Huang Tang high-dose group, liuwei yellow soup low-dose group, group administered30days, the rats to detect FBG, enzyme-linked immunosorbent assay (ELLSA) The serum cAMP, FINS levels and insulin resistance index (HOMA-IR), insulin sensitivity index (ISI); immunohistochemical staining was observed in liver p-PI3KP110p-Akt protein expression,; HE staining of pancreatic pathology, calculate islet (3cell function index (HOMA-β).Results:T2DM rats with FBG≥16.7mmol/L, the model group and the control group, serum FINS, cAMP levels and HOMA-IR values have increased, ISI, HOMA-β value decreased, the difference was significant (P<0.05); compared with the model group Liu Wei Di Huang Tang rat FBG low-dose group, cAMP, INS, HOMA-IR values decrease, ISI, HOMA-β value increased (P<0.05); compared with the control group, rat liver model group p-PI3KP110, p-Akt expression decreased (P<0.05); compared with the model group, Liu Wei Di Huang Tang low-dose group liver p-PI3KP110, p-Akt protein expression rose (P<0.05); rat pancreatic islet atrophy serious model group, reducing the number of islets islet irregular; Liu Wei Di Huang Tang rats increase in the number of pancreatic islets, islet shape relatively rules.Conclusion:High-sugar high-fat diet plus intraperitoneal injection of streptozotocin method can be successfully established rat model of type2diabetes and stable model; Liu Wei Di Huang Tang can lower fasting blood glucose in type2diabetic rats, decreased serum insulin, cAMP levels, improve insulin resistance; Liu Wei Di Huang Tang can protect islet structure to repair islet β-cell function; its mechanism of action may be related to regulating signal factor PI3KP110, Akt phosphorylation levels.
