The paper consists two parts:literature review and experimental research.First Section:Literature SummaryThere are two parts of literature summary. The first chapter is to introduce the research progress of Alzheimer’s disease,to explain the Alzheimer’s disease from neurotransmitter, neuroendoerine, immune system,and genetic factors.The second review mainly from traditional Chinese medicine on Alzheimer’s disease in history and discuss the pathogenesis, progress of modern research of acupuncture and moxibustion treatment of Alzheimer’s disease, including clinical research and experimental research on two aspects.Second Section:Experimental ResearchObjective:To observe the effects of SAMP8mice behavior indices, different effects of Electroacupuncture on SAMP8mice cerebral cortex APP,and APOE expression, to explore the mechanism of electroacupuncture treatment of senile dementiaMethods:Rats were randomly divided into seven groups:normal group, model group, drug group, acupuncture group, electro-acupuncture music group, the music group, non acupoint group, it is provided by experimental animal center of the First Affiliated Hospital of Tianjin College of Traditional Chinese Medicine. Totol in SAMP8mice and SAMR1mice were70.(1) using statistical to evaluate the effect of mice behavior indices;(2) Extracting cortical tissue from mice brain, and determinating the effects of different Electroacupuncture on SAMP8mice cerebral cortex APP and APOE expression by using Western Blog and immunohistochemistry.Results:1. Senile dementia’s mice after acupuncture treatment for15days, using the Morris’s test to detect the mice.The results (a) the experimental mice escape latency time and space exploration si a downward trend. On the third day model group compared with the control group, its space exploration time was delay in time, with statistical significance (P<0.01);(b) compared with the model group, the music group and non acupoint group space exploration time is decreased, but it was not statistically significant (P>0.05).(c) compared with the model group, the music electroacupuncture group, pulse electro acupuncture group and drug group of three groups of space exploration time are significantly reduced, with statistical significance (P<0.05).2. By using the Immunohistochemical method to respect the expression of senile dementia mice in brain cortex of APP and APOE.(a) The integral optical density and the number of positive cells in APP results. Compared with the normal group,the APP content of model group is more than normal group, there was a statistically significant difference (P<0.01); compared with the model group, there were no significant differences between the expression of non acupoint group and the music group in the APP (P>0.05); But the APP of EA group, drug group and the music electro-acupuncture group were obviously decreased, with statistical significance (P<0.01); compared the APP by music electro acupuncture group,electro acupuncture group and drug group, the expression of music electroacupuncture group had significantly decreased, with statistical significance (P<0.05).(b) The integral optical density and the number of positive cells in APOE results.Compared with the normal group, the APOE content of model group were significantly reduced, there is a statistically significant difference (P<0.01); compared with the model group, the expression of APOE in non acupoint group and music group has a rising trend but no significant difference (P>0.05); the expression of APOE with EA group, drug group and music electro acupuncture group in clear downward trend, with statistical significance (P<0.05,P<0.05, P<0.01); compared music electro acupuncture group and electro acupuncture group and drug group, the expression of music electroacupuncture group APOE has a clear upward trend, there is statistical significance (P<0.05).3. Western Blog to determine the expression of APP protein assay.(a) Compared with the normal group, the cortical APP protein bands of model group were obviously thickening deepened, with statistical significance (P<0.01); Compared with the model group, non acupoint group cortical APP protein band was no difference (P>0.05); And the music group cortex APP expression increased protein bands trend, but no statistical significance (P>0.05); EA group, drug group,music eletro acupuncture group cortical APP protein band expressed shallow downward trend, with statistical significance (P<0.05, P<0.05, P<0.01); Compared with EA group, the APP protein expression in cortical bands is obviously narrow and shallow, there was statistical significance (P<0.05). Western Blog to determine the expression of ApoE protein assay (b) Compared with the normal group, the cortical ApoE protein bands of model group were reduce and to pale, with statistical significance (P<0.01); With the SAMP8model group mice compared with non acupoint group mouse cortex, expression of ApoE had no significant difference (P>0.05); music group mice cortex ApoE expression has an upward trend, but no statistical significance (P>0.05); EA group, drug group,music eletro acupuncture group cortical ApoE protein band expressed deep and the upward trend, with statistical significance (P<0.05, P<0.05, P<0.01); And pulse electroacupuncture group contrast, music electroacupuncture group mouse cortical ApoE expression is significantly wider, become darker in color, with statistical significance (P<0.05).Conclusion:1. Behavioral detection the results of mice after the treatment with different electroacupuncture, it can improve the learning and memory ability of SAMP8miee,so that different acupuncture can certain therapeutic effect on senile dementia.2.Different effects of Electroacupuncture on expression SAMP8mice in APP and APOE model of the cortex play a regulatory effects of good, this kind of treatment can improve the learning and memory ability of mice, inhibit the prevention of senile dementia. To prove the mechanism of different electro acupuncture in the treatment of senile dementia, it can adjust the SAMP8mice model of neurotransmitter levels of monoamine.
