Common rush,which is the dried medulla of Juncus effuses L.,was a kind of common chinese medicinal herbr.There were more than100kinds of chemical compositions separated from it until now. Effusol,dehydroeffbsol and juncusol,which are the major phenanthrene in Juncus effuses L., have been conformed to exhibit anxiolytic effect by our previous study. In recent years,the demand of Juncus effuses L. increasing,but the wild plants were shorter than before,and also the normalized cultivation was getting less. So that,studying and making research of accumulation of phenanthrene content in Juncus effuses L.was very important for scientific cognition in different localities and the normalized cultivation.Juncus effuses L. was collected with2habitats in Jiangxi and Hunan province.Compared with the contents of active principle and soil elements in the two habitats,and discussed with influence on ecological factor to Juncus effuses L. phenanthrene.Provide scientific support on comprehensive quality analysis and evaluation.The vital significance of standardized production on Juncus effuses L.as the important meaning.which is in order to provide scientific basis for further expand the Jiangxi and Hunan province region to Juncus effuses L. development and utilization of quality resources.The dissertation included review section and experimental section.The review section was managed into two topics as following:1.the research status of Juncus effuses L.2.factors on the quality of herbal medicines.The experimental section included the development of HPLC method,the determination of the content of phenanthrenel and the study accumulation trend of phenanthrene in Jiangxi and Hunan province.1.A sensitive and specific HPLC method was developed and validated for the quantification of phenanthrene in Juncus effuses L. samples. A simple liquid-liquid extraction method with methanol was used to treat the Juncus effuses L. samples, then the samples were separated on an Agilent Eclipse-C18column by using methanol-water (57:43) as mobile phase at flow rate was1.0mL/min and UV detection at270nm. The precision and accuracy of the method were acceptable for Juncus effuses L analysis.2.Compared planting techniques between Jiangxi province and Hunan province.Juncus effuses L. was collected with the2habitats from2012and2013year.Using HPLC method for the determination of the content of effusol,dehydroeffbsol and juncusol.3.Copmared the content of phenanthrene in Jiangxi and Hunan province and study accumulation trend of phenanthrene in two habbits;the content of phenanthrene is significantly in different growing stages,phenanthrene content of Juncus effusus L. in Anren was same with the Juncus effusus L. in Fuzhou,dehydroeffusol content of Juncus effusus L. in Anren was significantly higher than the Juncus effusus L. in Fuzhou;Phenanthrene content of pith of Juncus effusus L was significantly higher than the same period of cortex;Planting Juncus effusus L. in the same field in2012and2013, phenanthrene in2013were significantly lower, Juncus effusus L. is not suitable for continuous cultivation.4. Using ICP-MS method for the determination of the content of P,K,Na,Ca,Fe,Mg,Mn,Cu and Zn elements in the two areas, using SPSS regression analysis, the calcium element content of root soil was the most important on form factor of Juncus effusus L.
