The whole thesis can be divided into three parts:source and evolution of Shenqi-Wan, the literature review of Zuogui Pili and Yougui Pili in the clinical application and experimental studies and the literature research of Multiple Sclerosis (MS) on the TCM diagnosis and intervention.On the source and evolution of Shenqi-Wan, there were three parts. The first part was based on the relevant content of Synopsis of Golden Chamber and the analysis of prescription compatibility, aiming to investigate thoroughly the original meaning of Shenqi-Wan The second part analyzed the dosage of prescriptions of during past dynasties, mainly including the dosage of Guizhi and Dihuang. The part aimed to investigate the evolution of Shenqi-Wan. The last part collected the prescriptions developed from Shenqi-Wan.On the literature review study, the article was based on the search of Zuogui Pili and Yougui Pili in the past five years and the summarization of its clinical application and experimental studies, aiming to analyze the clinical use range,main syndrome and the latest progress and research trend; the article analyzed TCM classification of MS according to clinical manifestation, and investigated the research status and development tendency of MS with the literature research findings.On the experimental research, the article investigated the effects of Zuogui Pili and Yougui Pili on differentiation of Thl7Cells and IL-23/IL-17axis of mice with experimental autoimmune encephalomyelitis(EAE)Objective:①To clarify the differences of Zuogui pill, Yougui Pill on therapeutic effect of EAE in mice.②To investigate the effects of Zuogui Pili and Yougui Pili on differentiation of Thl7Cells of mice with EAE.③To investigate the effects of Zuogui Pili and Yougui Pili on IL-23/IL-17axis of mice with EAE.Methods:EAE model was induced in C57BL/6mice, and were treated with Zuogui Pili, Yougui Pili,prednisone and saline, as Zuogui Pili group, Yougui Pili group, prednisone group and EAE group. Normal mice were selected as control group. Levels of IL-4, IL-6, IL-17, TGF-β in serum were detected by ELISA. Levels of IL-23p19, IL-17mRNA expression in brain tissue were detected by real time-PCR.Results:The incidence of EAE was significantly increased, while the neurological score and loss of weight were higher in Zuogui Pili, Yougui Pili and prednisone treated EAE mice than those in EAE group mice.Inflammatory cell infiltration in brain tissue was significantly decreased in Zuogui Pili, Yougui Pili and prednisone treated EAE mice than those in EAE group mice. IL-4, TGF-β levels in serum were significantly increased, while IL-6, IL-17levels in serum were significantly decreased in EAE mice than normal mice (P<0.05); IL-4, TGF-β levels in serum were significantly increased, while IL-6, IL-17levels in serum were significantly decreased in Zuogui Pili, Yougui Pili and prednisone treated EAE mice than those in EAE mice(P<0.05); IL-4,TGF-βlevels were decreased in Yougui Pili treated EAE mice than Zuogui Pili did(P<0.05).IL-23p19,IL-17mRNA levels in brain tissue were significantly decreased in Zuogui Pili, Yougui Pili and prednisone treated EAE mice than those in EAE group mice (P<0.01). IL-17mRNA levels was decreased in Yougui Pili treated EAE mice than Zuogui Pili did (P<0.01).Conclusions:Zuogui Pili and Yougui Pili play a preferable role in prevention and treatment of EAE mice, and the effort of Yougui Pili is slightly better than Zuogui Pili. Zuogui Pili and Yougui Pili may exert anti-inflammatory effect through inhibiting differentiation of Th17Cells, and the effort of Yougui Pili is better than Zuogui Pili. Zuogui Pili and Yougui Pili may exert anti-inflammatory effect through inhibiting IL-23/IL-17axis, and the effort of Yougui Pili is better than Zuogui Pili.
