Objective:To observe the effects of different doses of Jia Wei Dan Shen Yin preconditioning on myocardial cell apoptosis-related proteins during myocardial ischemia-reperfusion injury in rats.Methods:Sixty male rats were randomly divided into five groups:sham operation group,model group and JiaWeiDanShenYin low,medium,high dose groups.Rats in JiaWeiDanShenYin group were perfused with JiaWeiDanShenYin (3.02,6.04,12.08mg.kg"’)a week before operation,to the sham operation group and the model group,0.9%NS was given instead.The MIRI model was established by ligating left anterior descending coronary artery for30min and followed for90min.The activities of lactic dehydrogenase (LDH) and creatine kinase (CK) were detected using ELISA.Immunohistochemistry was used to detect myocardial tissue of rats,Bcl-2,Bax protein content.Results:Compared with the model group,JiaWeiDanShenYin pretreatment significantly suppressed the release of LDH and CK (Besides,the release of LDH and CK reduced along with increased dose.),the medium, high dose groups expression of Bcl-2protein were significantly with higher,while Bax in protein content were significantly with lower,the difference was statistically significant (P<0.05); compared with the low dose group,the expression of Bcl-2protein increased while Bax protein decreased significantly (P<0.05) during the mediun、 high dose groups,then between the two groups,Bcl-2expression in the high dose group compared to the medium dose group increased,Bax expression in the high-dose group compared to the middle dose group decreased,but no significant difference (P>0.05).Conclusion:JiaWeiDanShenYin pretreatment can regulation of myocardial tissue apoptosis and protect cardial tissue by increased Bcl-2expression and reduced Bax expression,furthermore presence of a dose-response relationship.
