Self emulsifying drug delivery systems (SEDDS) are isotropic mixures of poorly water-soluble drug, oil, surfactant and co-surfactant that can spontaneously form fine oil-in-water emulsion (most of the particles are less than5um) under the37℃gentle agitation in vitro or in vivo gastrointestinal peristalsis. The emulsion is thermodynamically stable, homogeneous, transparan and has small particle size.Numerous studies have confirmed that SEDDS have been utilized to improve the dissolution, absorption and bioavailability of poorly water-soluble drugs. The influence of different oil on SEDDS is compared in this study. Medium chain triglycerides, soybean oil, oleic acid monoglycerides are choosed as oil phase, then prepared the SEDDS of Andrographolide, Dehydroandrographolide, Potassium Dehydroandrograpolide Succinate formulation. Particle size is determinated by visual method and laser particle size scanner, which is common to evaluate The emulsion appearance. Results showed that emulsifying effect for chain triglyceride> oleic acid monoglycerides> soybean oil. This study compared the influence of different surfactant on SEDDS. Choose glycerine and1,2-propylene glycol as a surface active agent;, the SEDDS of Andrographolide, Dehydroandrographolide, Potassium Dehydroandrograpolide Succinate formulation is prepared. Particle size is determinated by visual method and laser particle size scanner, which is common to evaluate the emulsion appearance. Results showed that emulsifying effect for emulsifying effect as glycerin>1,2-propylene glycol. In this paper, we study the influence of different drugs structure that may affect on SEDDS. We choose andrographolide, Dehydroandrographolide, Potassium Dehydroandrograpolide Succinate as model drugs,.they have similar structure but different side chain groups, there are certain differences between physical and chemical properties. Prepared the SEDDS of Andrographolide, Dehydroandrographolide, Potassium Dehydroandrograpolide Succinate formulation. Particle size by visual method and laser particle size determination of scanner are common to compare emulsion appearance. Results showed that the emulsification effect of Andrographolide and Dehydroandrographolide andrographis is superior to Potassium Dehydroandrograpolide Succinate as a whole. This paper studied the influence of formulation of zeta potential, chose the prescription composition under different proportion and determine the emulsion particle size and zeta potential.the results showed that the prescription of drugs (including drugs) have an effect on the zeta potential. Also drug physical and chemical properties influence the zeta potential. The zeta potential and particle size are common to influence the stability of the emulsion. |
