Objective:Observe electro-acupuncture’s effect on chronic fatigue rat’s exhaustive swimming time、finding the platform time, spontaneous activity of24hours, supra chiasmatic neclue’s γ-aminobutyric acid’s expression and pineal gland’s melatonion positive cell expression’s circadian rhythms, to reveal the mechanism of that how electro-acupuncture adjusts chronic fatigue rat’s spontaneous activity’s circadian rhythms.Methods:Divide SD rats of clean grade to normal control group、model group、model electro-acupuncture group、model Sham injection group、model CRF injection group、 model CRF injection and electro-acupuncture group、model Antagonist-CRH injection group and model Antagonist-CRF injection and electro-acupuncture group randomly, make chronic fatigue model by chronic restraint and cold water swimming.Treat model Sham injection group、model CRF injection group、model CRF injection and electro-acupuncture group、model Antagonist-CRH injection group、model Antagonist-CRF injection and electro-acupuncture group with intracerebroventricular administration of surgery separately,while treat model electro-acupuncture group、 model CRF injection and electro-acupuncture group、model Antagonist-CRF injection and electro-acupuncture group with electro-acupuncture on BL23、ST36.Observe and record the exhaustive swimming time、finding the platform time and spontaneous activity of24hours of all groups.Observe and record supra chiasmatic neclue’s γ-aminobutyric acid’s and melatonion receotor positive cell’s expression at0:00,4:00、8:00、12:00、16:00、20:00by immunohistochemistry.Results:1.Exhaustive swimming time and finding the platform time after treatment:exhaustive swimming time after treatment,model group compared with normal control group time to be shortened,finding the platform time conversely.Exhaustive swimming time of model electro-acupuncture group extended compared with model group.finding the platform timeconversely. Exhaustive swimming time of model CRF injection group、model CRF injection and electro-acupuncture group after treatment extended obviously compared with model group、model Sham injection group(P<0.05).finding the platform time conversely.Exhaustive swimming time after treatment,model Antagonist-CRH injection group、model Antagonist-CRF injection and electro-acupuncture group compared with normal control group%model electro-acupuncture group、model CRF injection group、model CRF injection and electro-acupuncture group time to be shortened, finding the platform time conversely.2.Spontaneous activity after treatment:①Model group’s peak value came obviously later compared with normal control group (P<0.05); Model electro-acupuncture group’s peak value came obviously sooner than that of model group.Model CRFH injection group and model CRF injection and electro-acupuncture group’s peak value came obviously sooner than that of model group、model Sham injection group.(P<0.05); Model Antagonist-CRF injection group’s peak value came obviously later compared with normal control group、model group、model electro-acupuncture group、model Sham injection group、model CRF injection group、model CRH injection and electro-acupuncture group (P<0.05); Model Antagonist-CRF injection and electro-acupuncture group’s peak value came obviously later compared with normal control group、model electro-acupuncture group、model CRF injection group and model CRF injection and electro-acupuncture group.②The daytime activity of model group increased compared with normal control group (P<0.05); the daytime activity of model electro-acupuncture group increased compared with model group (P<0.05).The daytime activity of model CRF injection group and model CRF injection and electro-acupuncture group reduced compared with model group and model Sham injection group.The daytime activity of model Antagonist-CRH injection group increased compared with normal control group、model electro-acupuncture group、model CRF injection group and model CRF injection and electro-acupuncture group.The daytime activity of model Antagonist-CRF injection and electro-acupuncture group increased compared with normal control group、model electro-acupuncture group and model CRF injection and electro-acupuncture group.③The activity at night of model group reduced compared with normal control group;the activity at night of model electro-acupuncture group increased compared with model group.The activity at night of model CRF injection group and model CRF injection and electro-acupuncture group increased compared with model group and model Sham injection group.The activity at night of model Antagonist-CRH injection group and model Antagonist-CRF injection and electro-acupuncture group reduced compared with normal control group、model electro-acupuncture group、model CRF injection group and model CRF injection and electro-acupuncture group.Supra chiasmatic neclue’s γ-aminobutyric acid’s expression after treatment:There is circadian rhythms in the expression of model electro-acupuncture group、model CRF injection group and model CRF injection and electro-acupuncture group(P<0.05); while there is no circadian rhythms in the expression of model group、model Sham injection group、model Antagonist-CRH injection group and model Antagonist-CRF injection and electro-acupuncture group (P>0.05).3.Optical density of pineal gland’s melatonion positive cell expression:①There is circadian rhythms in the expression of model electro-acupuncture group、model CRF injection group and model CRF injection and electro-acupuncture group (P<0.05); while there is no circadian rhythms in the expression of model group、model Sham injection group、model Antagonist-CRH injection group and model Antagonist-CRF injection and electro-acupuncture group (P>0.05).②Optical density of pineal gland’s melatonion positive cell expression at0:00of model Antagonist-CRH injection group and model Antagonist-CRF injection and electro-acupuncture group decreased obviously compared with model group (P<0.05); Optical density of pineal gland’s melatonion positive cell expression at8:00ofmodel Antagonist-CRF injection and electro-acupuncture group decreased obviously compared with normal control group、model electro-acupuncture group、 model CRF injection group and model CRF injection and electro-acupuncture group,, there were significant statistical differences(P<0.05).Conclusion:Chronic restraint and cold water swimming caused rats’s circadian rhythms loss of supra chiasmatic neclue’s γ-aminobutyric acid’s expression and pineal gland’s melatonion positive cell expression,activity unusual during the daytime and night and obvious physical fatigue and mental fatigue.More content of hypothalamic paraventricular nucleus CRH benefits electro-acupuncture on BL23, ST36’s effect on recovering chronic fatigue rat’s supra chiasmatic neclue’s γ-aminobutyric acid and pineal gland’s melatonion’s circadian rhythms,adjusting activity during the daytime and night,improving fatigue and vice versa.Hypothalamic paraventricular nucleus CRH is the key to electro-acupuncture’s effect on adjusting chronic fatigue rat’s circadian rhythms and improving fatigue.
