AIM:To study the effect of compound buyang huanwu tang modification on proliferation of colorectal cancer cells as well as growth of subcutaneous xenograft in nude mice, so as to initially elucidate the mechanism; To investigate the clinical effects of buyang huanwu tang in combination with chemotherapy through observing the physical conditions-clinical symptoms, level of tumor markers, short-term objective response and extent of reaction of side-effects in patents。METHODS:Cell culture, light microscopy and enzymatic histochemistry technique were used to observe the effect of compound buyang huanwu tang modification on proliferation of human coiorectal cancer cell strain LOVO。Load tumor animal model were established by vaccination of human colon cancer cell LOVO in nude mice。 Then model group, low-dose Chinese medicine group and high-dose Chinese medicine group were set up。 ELISA was used to examine the quantity of CEA, and compared each groups on the general status, ponderal growth, tumor variation and quantity of CEA in serum。HE-staining for property, data processing by statistical analysis。The mechanism that antitumor effects of buyang huanwu tang modification in colorectal cancer was initial elucidated。45patients with advanced colorectal carcinoma were randomly divided into standard chemotherapy group and treatment group which meant combination of buyang huanwu tang and chemotherapy。To observe and compare the differences about physical conditions, clinical symptoms, short-term objective response, level of tumor markers and reaction of side-effects in patents。 All results were processed by statistical analysis。RESULT:It was indicated that buyang huanwu tang modification could inhibit the proliferation in colon cancer cell LOVO, which was in proportion to drug concentration; There was no obvious change between two groups under the observation of light microscopy; The size and weight of tumor in treatment group were significantly minor than that of control group, meanwhile there was no significant difference in serum CEA quantity between two groups; As to clinical study, treatment group tend to show better improvement than control group in aspect of physical conditions, clinical symptoms, level of tumor markers and reaction of side-effects in patents。CONCLUSION:Buyang huanwu tang modification may play a role in inhibition to the proliferation of colorectal carcinoma cells in vitro, and suppress the growth of transplanted tumor in nude mice; As to clinical observation, buyang huanwu tang modification may play a role in the improvements in aspect of patient’ physical conditions, clinical symptoms, tumor markers level and response of chemotherapy。... |
