Objective:By observing the miao medicine Rekechuan capsule on MMP-9gene expression in the rat model with bronchial asthma airway, further explore MMP-9gene regulation effect on bronchial asthma airway remodeling and hot capsule treating bronchial asthma in the molecular biology mechanism, which might provide theoretical basis for the research and development of new drugs for the hot cough and gasp the capsules.Met:80male SD rats were randomly divided into normal group (group A), miao medicine group (B group), model group (group C) and dexamethasone group (group D), which means20rats each group. All the groups were made into bronchial asthma model,except the normal group. Give mould with physiological saline to group A and the rest of the group of rats with10%egg albumen (OVA)100mg, AL(OH)3100mg mixed suspension.inject in the abdominal to get Sensitization model On the4th day and11th day. From the18th day, make group A inhale aerosol of normal saline, the rest of the group with1%OVA solution atomization for2weeks, half an hour each time.lavage the group A and group C with amount of normal saline, group B with the capsule powder, inject group D with intraperitoneal of dexamethasone acetate injection half an hour before atomization.Excute all the groups of rats within6hours and take lung tissue and airway specimens which would be HE stained, and observed the airway inflammation and through immunohistochemistry and RT-PCR technique, MMP-9and MMP-9mRNA was detected in the rat model with bronchial asthma in the airways of the expression level in order to know the influence of the capsule.Results:The thickness of bronchiole smooth muscle:The thickness obviously increased in group A group B and group D. The difference is very statistically significant that bronchiole smooth muscle in group C is thicker than group A(P<0.01). The difference is very statistically significant that bronchiole smooth muscle in group B is thinner than group C(P<0.01). The difference is very statistically significant that bronchiole smooth muscle in group D is thinner than group C(P<0. 01). There is no significant difference of the thickness of bronchiole smooth muscle between group B and group D.The expression of MMP-9and MMP-9mRNA:the level of the expression is the lowest in group A. The difference is very statistically significant that the level of the expression in group C is higher than group A(P<0.01). The difference is very statistically significant that the level of the expression in group B is lower than in group C(P<0.05). The difference is very statistically significant that the level of the expression in group D is higher than group C(P<0.01).And there is no significant difference of the expression of MMP9between the miao medicine and dexamethasone in the two groups.Conclusion:The thickness of the bronchiole smooth muscle in group B and group D was thinner than group C,which means the miao medicine Rekechuan capsule can inhibit the smooth muscle hyperplasia.The expression level of MMP-9、MMP-9mRNA in the bronchiole of asthma rats is obviously higher than the normal rats,which means MMP-9is a cause of the airway remodeling.The expression level of MMP-9、MMP-9mRNA in the bronchiole in group B and D is obviously lower than group C,which means the Rekechuan capsule plays an important role of preventing the airway remodeling by controlling the expression of MMP-9.
