OBJECTIVE:To observe the effect of traditional Chinese Medicine formula JianPi Shu Gan Decoction(JPSGD) on hepatic fibrosis in mice caused by CCl4. Andcompare its effect with that of traditional Chinese Medicine formula Huo XueHua Yu Decoction(HXHYD). And at the same time, to explore its possible mechanism.METHOD:280malecleangradeC57BL/6Jmicewererandomlydividedinto4groups:the normal control group, the model control group, the Jian Pi Shu Gan groupand the Huo Xue Hua Yu group, in which there were70mice. Except the normalcontrol group, other3groups which included the model control group, the JianPi Shu Gan group and the Huo Xue Hua Yu group, were abdominally injected with20%CCl4olive oil solution at a dose of1ml/kg body weight twice a week foreight weeks. And the normal control group was abdominally injected with100%olive oil at a dose of1ml/kg body weight twice a week for eight weeks. Sincethe fifth week, the Jian Pi Shu Gan group and the Huo Xue Hua Yu group wereintragastricly administered Jian Pi Shu Gan Decoction and Huo Xue Hua YuDecoction respectively at a dose of0.2ml/10g once every day for4weeks. Micein the normal control group and the model control group were given the same volumeof deionized water in oral way. After the end of experiment, the levels of livertissue matrix metalloproteinases (MMP-1), the inhibitor of matrixmetalloproteinases(TIMP-1), serum hyaluronic acid (HA), laminin protein (LN),procollagen III (PC Ⅲ) and collagen IV (Ⅳ-C) were detected by Enzyme-linkedimmunosorbent assay. Meanwhile, histological changes in mice liver pathologywere observed by light microscopy.80male clean grade C57BL/6J mice wererandomly divided into4groups:the normal control group, the model control group,the Jian Pi Shu Gan group and the Huo Xue Hua Yu group, in which there were20mice. Except the normal control group, other3groups which included the modelcontrol group, the Jian Pi Shu Gan group and the Huo Xue Hua Yu group, were abdominally injected with20%CCl4olive oil solution at a dose of1ml/kg bodyweight twice a week for eight weeks. And the normal control group was abdominallyinjected with100%olive oil at a dose of1ml/kg body weight twice a week foreight weeks. Since the fifth week, the Jian Pi Shu Gan group and the Huo XueHua Yu group were intragastricly administered Jian Pi Shu Gan Decoction and HuoXue Hua Yu Decoction respectively at a dose of0.2ml/10g once every day for4weeks. Mice in the normal control group and the model control group were giventhe same volume of deionized water in oral way. After the end of experiment,the levels of liver tissue matrix metalloproteinases (MMP-1), the inhibitorof matrix metalloproteinases (TIMP-1), serum hyaluronic acid (HA), lamininprotein (LN), procollagen III (PC Ⅲ) and collagen IV (Ⅳ-C) were detected byEnzyme-linked immunosorbent assay. Meanwhile, histological changes in miceliver pathology were observed by light microscopy.RESULT:The levels of liver tissue TIMP-1, HA, LN, PCⅢ, Ⅳ-C of the Jian PiShu Gan Group and Huo Xue Hua Yu group were all significantly lower than thoseof the model group (P<0.05). The levels of TIMP-1, HA, LN, PCⅢ, Ⅳ-C of theJPSG group were significantly lower than those of the HXHY group (P<0.05).Compared with the normal group, the MMP-1levels of other three groups were allsignificantly higher (P<0.05). However, the serum MMP-1levels of the threegroups have no significant difference (P<0.05).CONCLUSION:Jian Pi Shu Gan Decoction shows a striking effect of inhibitionand treatment to hepatic fibrosis in mice caused by CCl4.
