Obiective:This investigation was performed to develop a method to increase the content of salidroside in extract of Fructus Ligustri lucidi using acid hydrolysis.Through the comparison research of activity of anti-osteoporosis of crude and prepared Fructus Ligustri lucidi, it provided theoretical basis of anti-osteoporosis effect of prepared Fructus Ligustri lucidi in clinical application.Methods:With the content of salidroside and tyrosol as index, the thogonal design was adopted to acquire the optimum technology for the extraction of salidroside by investigating the feed liquid ratio, hydrolysis time and hydrolysis temperature on the influence of the content and optimum extraction and purification process conditions using HPLC.The method for the simultaneous quantitative analysis of salidroside, hydroxytyrosol, and specnuezhenide of Fructus Ligustri lucidi was developed. Then ALP activity was used to study its impact on UMR-106cell differentiation. Using liposome transient transfection method and the transfection of ERβ/β expression plasmid in HeLa, it was observed that activation of reporter gene transcription of ERE in crude and prepared Fructus Ligustri lucidi and plant estrogen-like effect. It was examined that the total antioxidant capacity size using DPPH and ABTS and FRAP.Results:The feed liquid ratio and hydrolysis time played the leading role. The optimum process conditions were in fructus ligustri lucidi concentrate adding volume ratio of20:1glacial acetic acid and backflowed20hours in100℃. The content of salidroside by hydrolysised was about three times as much without acid hydrolysised. Fructus ligustri lucidi raw material was added amount of6times water by reflux extraction for three times, each time for1hour. D4020type resin column purification in fructus ligustri lucidi process conditions for:the sample concentration was0.5g·mL-1extract, feed liquid and macroporous resin ratio is1:30, adsorption velocity for1BV·V-1, with20%ethanol elution5times resin volume. It was higher that the content of salidroside, hydroxytyrosol, and specnuezhenide of prepared Fructus Ligustri lucidi than raw products. Before and after processing can induce activation of reporter gene transcription of ERE in different degrees and promote the UMR-106cells increased. The effct of prepared Fructus Ligustri lucidi is better than that of raw products.The total antioxidant capacity of prepared Fructus Ligustri lucidi precede the raw product.Conclusion:By optimizing the extraction and purification technology,the ice acetic acid hydrolysis method can significantly raise the content of salidroside in Fructus Ligustri lucidi.The prepared Fructus Ligustri lucidi has the effect of anti-osteoporosis and superior to the raw product.
