Purpose: to investigate the curative effect of traditional Chinese medicine Huangdijixiantangand the influence of Bcl-2/Bax pancreatic isleta β cells on arts with streptozotocin (STZ)-induceddiabete.Materials and methods: use the high glucose and lipid diet plus stress stimulate and singletime little dose intraperitoneal injection of STZ to make the diabetic type2model of rats. Andthen treat with traditional Chinese compound Huangdijixiantang and diformin separately. Aftercure for2or4weeks examine the weight and blood glucose and adjust the dose of medicineaccording to the measure. The eighth week execute those model rats, examine weight and bloodglucose, collect sample and observed them under the HE dyeing, TUNEL andImmunohistochemical dyeing to measure tissue morphostructure. Estimate the function andmorphostructure of β-cell, observe the opoptosis and the expression of Bcl-2and Bax.Results:1The end of the experiment, Huangdijixiantang and metformin able to lower the bloodglucose in the diabetes type2model of rats.(P<0.05)2Morphological observation: After treatment, the normal group rat pancreatic exocrinevisible islet distribution, islet island-shaped rounded and complete, the island cell body is full,arranged in dense; the model rats islet volume was significantly smaller, irregular contour, Islandwithin the cells decreased, disorganized; similar to the performance of the low dose group andmodel group; Chinese medicine, high-dose treatment group compared with metformin isletisland-shaped full round, the number of cells on the island than in model group increased, closelyarranged in uniform.3Islet cell apoptosis Index: model group was significantly higher than the normal group (P<0.05); Chinese medicine, middle dose group, western medicine group apoptotic index wassignificantly lower than the model group (P <0.05), traditional Chinese medicine, low-dose groupapoptosis index with the model group was no significant difference (P>0.05).4Bcl-2, Bax protein expression: After treatment, comparison group of Bcl-2positive cellsaverage, model group was significantly lower than normal group (P <0.05); low dose group andthe model of Bcl-2positive cells no difference (P>0.05) between the number of high, middle dose group; Chinese and western medicine group BCI-2positive cells was significantly higher than themodel group (P <0.05). Bax positive cells of the model group was significantly higher than thenormal group;(P <0.05) each traditional Chinese medicine, the middle dose group and westernmedicine group and normal group Bax positive cells was significantly lower than the model group(P <0.05); low dose of Bax positive cells was significantly higher than that of traditional Chinesemedicine, middle dose group (P <0.05)5(Bcl-2\Bax) ratio comparison: model group was significantly lower than normal group(P <0.05); no difference between the low dose group and model group (P>0.05); Chinesemedicine, middle dose group, Western medicine group (Bcl-2\Bax) ratio was significantly higherthan the model group (P <0.05),Conclusion:Huangdijixiantang able to lower the blood glucose in the diabetes type2modelof rats and can inhibit the Islet β cell apoptosis, which may be up-regulated the expression ofBcl-2and inhibit the expression of Bax.
