Objective:Screening and identification of the genes related to osteosarcomaļ¼Œand furtherrevealing the molecular mechanism of Osteosarcoma.Methods:Surgically resected fresh osteosarcoma tissue and normal tissue used forcomparison from3patients with osteosarcoma were selected for this study.The totalRNA of the tissus were extracted for reverse transcription,then marking andhybridization with Affymetrix gene chip.Empirical bayes method was used to screenthe Differentially Expressed Genes in human osteosarcoma.Next the Gene Ontologynomenclature was used to dealt with the Differentially Expressed Genes.Alsoenrichment analysis was carried out to study the molecular function, biologicalprocesses, and signaling pathways of them.then,we selceted3genes.Finally, weverified part of the gene expression by RT-PCR and Western blot.Results:Of105differentially expressed genes releated to Osteosarcoma which wereScreened by Gene chip,42genes were up-regulated and63were down-regulated.Theirmain molecular function was Protein binding, etc.Biological pathways was immuneresponse, complement activation and signal transduction, etc.And the mainly involvedsignaling pathways was immune response, cell viability, cell movement and negativeregulation of cell apoptosis, etc.BAX, HMMR and SHB were varified by RT-PCR andWestern blot.And the results were in conformity with the chips.Conclusion:The abnormal expressions of BAX, HMMR and SHB are related to the onsetof osteosarcoma.
