Part1Study of MWM Spatial Cognitive Behavior[Objective] Establish the model of pure cerebral concussion (PCC) and multiple cerebral concussion(MCC) in rats, then observe the changes of MWM test post PCC and MCC in rats so as to evaluate their spatial cognitive function. Thus it will provide references for diagnosis and treatment to clinical cerebral concussion(CC) syndrome patients.[Methods]1Animal group:40adult healthy SD rats were chosen, male and female, weighting260-280g. Rats were randomly assigned into the Control group(n=10)、PCC、MCC24h、MCC72h injury groups(n=10for each).2PCC、 MCC24h、MCC72h injury model was produced in rats using a simple pendulum device.3Using the MWM test to evaluate the spatial cognitive function of rats. Including16days of under-water platform test,2days of upper-water platform test, and1day no platform test.[Results]1In the under-water platform test, the escape latency(time to find the platform) of Control group decreased quickly, and then stabilized, while the escape latency of injury groups decreased quickly, rebounded in about8d, and then stabilized. Compared to the Control group, the escape latency of injury groups significantly increased,1-4d of the PCC group,1-16d of the MCC24h group, the rest13days of the MCC72h group except7d,10d,15d were statistically significant(P<0.05). Compared to the PCC group, the escape latency of the MCC groups significantly increased,1-16d of the MCC24h group,3d、13d of the MCC72h group were statistically significant(P<0.05). Compared to the MCC72h group, the escape latency of the MCC24h group significantly increased,1-14d were statistically significant (P<0.05).2In the upper-water platform test, the escape latency of the MCC groups significantly increased.3In the no platform test, compared to the other groups,the time remaining in the original platform quadrant of MCC24h group was significantly reduced.[Conclusion]1Different types of cerebral concussion have significant damaging effects on the spatial cognitive function in rats.2Comparing to the PCC, MCC does more damaging effects on spatial cognitive function in rats.3It shows that extending the injury interval, may be effective to reduce the SIS damaging effects on spatial cognitive function of MCC in rats. Part2Study of The Expression of GABABR2and VGAT[Objective] Establish the model of pure cerebral concussion (PCC) and multiple cerebral concussion(MCC) in rats, then quantitative analysis the expression of GABABR2and VGAT in each cerebral region after injury by Western-Blot analysis. Thus it will provide references for diagnosis and treatment to clinical cerebral concussion(CC) syndrome patients.[Methods]1Animal group:152adult healthy SD rats were chosen, male and female, weighting260-280g. Rats were randomly assigned into the Control group (n=8)、PCC、MCC24h、MCC72h injury groups(n=48for each).2PCC、MCC24h、 MCC72h injury model was produced in rats using a simple pendulum device.3Quantitative analysis the expression of GABABR2and VGAT in frontal cortex, brainstem, hippocampus, basal ganglia and septum1d,2d,4d,8d,16d,24d after injury with the application of Western-Blot experiment.[Results]1GABABR2:Compared to the Control group, the expression of GABABR2in injury groups were significantly decreased in all cerebral regions Id,2d,4d,8d,16d,24d after injury, and each time point were statistically significant(P<0.05). The expression of GABABR2declined rapidly at first, and reached a minimum at8d, then slowly raise. There were significant differences between the PCC, MCC24h, MCC72h group in each cerebral region. The MCC24h group decreased the most, then the MCC72h group followed by, and the PCC group shown the smallest decrease.2VGAT:Compared to the Control group, the expression of VGAT in injury groups increased rapidly at first, and reached a peak at8d, then slowly declined in all cerebral regions, and2d、4d、8d after injury were statistically significant(P<0.05). There were significant differences between the PCC, MCC24h, MCC72h group in each cerebral region. The MCC24h group increased the most, then the MCC72h group followed by, and the PCC group shown the smallest increase.[Conclusion]1Different types of cerebral concussion can reduce the expression of GABABR2, while increase that of VGAT in each cerebral region after injury, this may play an important rule in the development process of secondary cerebral injury.2 In hippocampus and the prefrontal cortex which closely related to spatial cognitive function of rats, the lowest point of GABABR2’ expression after injury appeared at8d, while the peak of VGAT’ expression after injury appeared at4d.3Comparing to the PCC, MCC does more damaging effects on spatial cognitive function in rats. It shows that extending the injury interval, may be effective to reduce the SIS damaging effects on spatial cognitive function of MCC in rats.
