Complement C3, C5 Expression Analysis In The Jaw And Limb Regeneration Under The Yunnan Blue-tailed Salamander Process

[Objectives]To investigate complement C3and C5expression in mandibular and forelimb regenerative tissue with a local species-Yunnan blue tail newt (Cynops cynaurus).[Methods] Sixty wild type adult Cynops cynaurus were divided into two groups of30randomly. One third amputations in the middle of mandible or right forelimb amputations were performed separately in the consistent groups under general anesthesia of Tricaine. The regenerative tissues of jaw and right forelimb as well as normal tissue of jaw and contralateral left forelimb were harvested respectively on the seventh and twenty-first days after amputation. The expression level of complement C3and C5was examined by RT-PCR and immunohistochemistry. The expression and distribution of complement C3and C5in regenerative tissues during different period were analyzed by in immunohistochemistry analysis.[Results] All newts survived and jaws and limbs regenerated partially without any complication during these short experimental periods. The expression level of complement C3and C5was examined by RT-PCR.Epimorphosis of partial jaw and right forelimb within experimental period; The complement C3and C5were expressed in the regenerative tissues of mandible and right forelimb regeneration, but not in the normal tissue of mandible and Contralateral left forelimb. C3expressed mainly in the regenerative tissues of jaw and right forelimb blastema and the wound epithelium exclusively,C5in the regenerative tissues of mandible and right forelimb wound epithelium exclusively but not expressed in blastema. Complement C3, C5expression intensity changes enhanded with time change.[Conclusions]1、The expression level of complement C3and C5was examined by RT-PCR.2、It seemed that the regeneration of mandible and right forelimb were associated with complement C3and C5.3、The complement C3and C5were expressed in the regenerative tissues of mandible and right forelimb, but not in the normal tissues of mandible and Contralateral left forelimb.C3expressed mainly in the regenerative tissues of mandible and right forelimb blastema and the wound epithelium exclusively,C5in the regenerative tissues of mandible and right forelimb wound epithelium exclusively but not expressed in blastema.
