Objective To observe the expression of RUNXl duirng the process of mandibularfirst molar and incisor of Balb/c mice and explore the function of Runt-relatedtranscription factor in tooth development.Method Forty Balb/c mice were randomly separated into5groups according to theexpeirmental time of tooth development (E19.5d, POd, P6d, P14d and P28d). Theincisor section and first molar section of the mandible were then removed andisolated. The specimens were fixed in4%paraformaldehyde in O.IM phosphate for24h and decalcified in10%ethylene diaminetetraacetic acid at4°C for certain daysdepends on the experimental time. After being dehydrated, cleared in xylene, andembedded in paraffin blocks,5|im eiral sections were cut parasagittally. HE andImmunohistochemical staining were carried out to detect the expressions of RUNXLResults RUNXl was detected both in first molar and incisor at El9.5day. And thenprogressively increased at PO,6,14,28days in first molar. While in the developmentof mice incisor, the expression of RUNXl increased at PO and P6days but showed adecreased trend at P14and P28days. Conclusions The expression pattern of RUNXl during the process of mandibularincisors between mandibular first molars of Balb/c mice were observed different. Itcan be speculated that RUNXl participates in differentiation and maturation ofameloblast and odontoblast and expresses tissue-specific. A peakpoint was detected at P6day, and a downward trend was observed atP14day and P28day.Conclusions The expression pattern of RUNX2during the process of mandibular incisors between mandibular first molars of Balb/c mice shared many similarities aswell as differs. Both of the experiments turned out that RUNX2mainly regulates thematuration of dentin and the process of biological mineralizing. Itcan be speculated that RUNXS might be related with the deposition of predentin andformation of dentin.
