| Kelp is a natural healthy food and contain a variety of valid compositions with definite nutritional function. At the moment, the scholars in the world focus on a particular of a variety of valid compositions in kelp, lack of comprehensive utilization of kelp. In this paper, using a low-temperature-microwave extraction technology, to extraction and separation a variety of valid compositions in kelp, to lay the technical foundation for the dynamic-microwave extraction(DME) with kelp.The following sections was research and results:(1) Extract conditions of the static state-microwave extraction(SME) in a small test were optimized by single factor method and L9(34) orthogonal experimental design method. The experiments shows that85%ethanol was as solvent, extraction temperature60℃, microwave power300W, solid-liquid ratio1:6→1:4, exteaction time20min, the ratio of extraction is38.59%. To separation the kelp extraction, the kelp low polar components was3.4%, the kelp polyphenol content was14.23(phloroglucinol)mg/g.(2) The emulsification process was core material13.2%, β-cyclodextrin19.9%, casein0.7%, emulsifier2.6%; The conditions of spray drying was inlet air temperature170℃, the charging rate13.95mL/min, fan speed4.7m3/min, the rate of retention95.23%, moisture content was3.35%, the yield was74.88%. the power was celadon and nonhygroscopicity.(3) The preferred method for isolation and purification of mannitol in kelp, the ratio of superior sea salt was30.1%, the ratio of coarse manntiol was20.7%, the purity of coarse manntiol was91.9%. To purification of coarse manntiol, the ratio of manntiol was82.8%, the purity of coarse manntiol was98.1%.(4) Using single factor method and L9(34) orthogonal experimental design method to extracted the polysaccharide in kelp. The experiments shows that water was as solvent, extraction temperature75℃, microwave power500W, solid-liquid ratio 1:30, exteaction time20min, the ratio of extraction is11.3%, the purity of the polysaccharide in kelp was16.57%.(5) Using single factor method and L9(34) orthogonal experimental design method to extracted the sodium alginate in kelp. The experiments shows that0.10mol/L sodium carbonate was as solvent, extraction temperature50℃, solid-liquid ratio1:30, exteaction time20min, the ratio of extraction is15.43%, the viscosity of the sodium alginate in kelp was392mps.(6) The detection method is simple and rapid, low-cost, accurate and is readily the producion process, and it can be utilized in the quantitative determination and product standarization.This paper introduced low temperature-dynamic microwave technology, research and development of high-quality, low-cost extract products to increase the added value of kelp, to improve the comprehensive utilization of kelp. |
