Objective: To study the protection of n-Butanol extract on the damage model of vascular endothelial cell induced by H2O2, through the expression of related gene and signal transducion.Methods: human umbilicus vascular endothelial cells (HUVEC) were cultivated as usual. The damage model of HUVEC was induced by 100μmol/1 H2O2. 5 groups were divided: there are control group, damage group, n-Butanol extract groups in high, medium and low dosage. Each groups were cultured for 24 hours, then cells were collected. The morphology were observed by flueroscope, the proliferation of HUVEC were tested by MTT assay, and the expression of related gene (bcl-2、VEGF、c-fos) and pathways of signal transduction (CREB、NF-KB、P38-MAPK) were determined by immunohistochemistry staining and flow cytometer.Result and Conclusions: n-Butanol extract could enhance proliferation of HUVEC and protect its damage by H2O2 by lowering cell apoptosis and increasing cells in S cycle, and regulating the expression of related gene: bcl-2、VEGF、c-los and pathways of signal transducion: CREB、NF-KB、P38-MAPK.
