Objective: The protective effect and mechanism of Carthamus tinctorius L. injectionin the intervention of skin flap ischemia reperfusion injury are studied.Methods:80healthy adult SD rats were randomly divided into sham operation group(SO group), ischemia reperfusion group (IR group), Carthamus tinctorius L. injectiongroup (HHI group) and dextran-40group (D-40group). Each group of rats wererandomly divided into ischemia for2h,4h,6h,8h and1H reperfusion group, a total of16teams. In the30min before experiment modeling each group was respectivelyintraperitoneal medicined according to the specified drug. SO group:2ml/Kg normalsaline; IR group:physiological saline2ml/Kg; D-40group:8.5ml/Kg; HHI group:2ml/Kg. After operation, the skin flap color of every group rats was observed, histologystructure of the skin flap was observed under the light microscope, ultrastructuralorganization of the skin flap was observed under the transmission electron microscope,hemorrheology indexes were tested, the SOD activity and MDA content of skin tissueand serum of every group rats were measured.Results: Under general observation, the skin flap reperfusion swelling in HHI groupand D-40group than in IR group was obviously lighter, the skin flap color wasobviously ruddyer; under the light microscope and the transmission electron microscope,the extent of tissue edema and infiltration of inflammatory cells was smaller, the injurydegree of organelle and the internal elastic lamina were lighter; HHI group and D-40group ischemia for6h,8h and1H reperfusion group compared with IR group: SODactivity in skin flap and serum was higher, with statistical significance (P <0.05); MDAcontent in skin flap and serum was lower, with statistically significant (P <0.05); Lowand high shear rate blood viscosity, and erythrocyte aggregation index values are small,with statistically significant (P <0.05). Conclusion: Carthamus tinctorius L. injection can reduce the infiltration ofinflammatory cells to the organization and the skin flap of tissue edema; Carthamustinctorius L. injection can eliminate oxygen free radicals in the skin flap tissue, protectsuperoxide dismutase activity, reduce lipid peroxide bringing, decrease MDA content,simultaneity can prevent or alleviate the injury of vascular endothelial cells; Carthamustinctorius L. injection can improve or interven the economy systemic blood rheology,that is validated by experiment.
