AIMUsing different concentrations of ethanol intervention microglia to choosing the suitable concentration of ethanol, which can make the microglia damage instead of a large number of death;To investigate the effects of HU-308on secretion of IL-6in alcohol-induced microglia.MethordsRat microglia culture was exposed to alcohol (0.00%,0.025%,0.050%,0.1%,0.2%) in vitro. Then IL-6levels are detected by Elisa method after72hours.The concentration of0.050%was selected for subsequent concentration after experiments and statistical analysis.When the microglia of mice grow to cover80%of six orifice bottle, using0.050%ethanol intervention it. Microglia were divided into control group, lumol/LHU-308intervention group,5umol/LHU-308intervention group and10umol/LHU-308intervention group. Using lumol/L,5umol/L,10umol/LHU-308intervention microgliam,respectively.IL-6levels detected by Elisa method after72hours. Analysis the collected data and draw conclusions.ResultsThere is not significant difference between0.00%ethanol and0.025%ethanol group.There is significant difference between0.00%ethanol and0.050%ethanol group and between0.00%ethanol and0.10%ethanol group. There is significant difference between0.025%ethanol and0.050%ethanol group and between0.025%ethanol and0.10%ethanol group. There is not significant difference between0.050%ethanol and0.10%ethanol group.In the control group, lumol/LHU-308intervention group,5umol/LHU-308intervention group and10umol/LHU-308intervention group,the average number of IL-6decreased sequentially.There is statistically significant difference between the intervention group and lumol/LHU-308control group,5umol/LHU-308intervention group,10umol/LHU-308intervention group.There is statistically significant difference between1umol/LHU-308intervention group and5umol/LHU-308intervention group,10umol/LHU-308intervention group.There is not significant difference between5umol/LHU-308intervention group and10umol/LHU-308intervention group.Conclusion0.025%ethanol has no obvious influence on microglia secretion level of IL-6.0.20%ethanol can make most of microglial cells to die.0.050%ethanol and0.10%ethanol treatment has no obvious difference.Using0.050%ethanol concentration for follow-up experiments. HU-308may reduce the level of IL-6secreted by microglia.In a certain concentration range, the level of IL-6decrease with the increased concentration of HU-308.
