Objective:Uterine leiomyoma is the most common gynaecological benign tumour and has make a great effect on reproductive health and well-being, which is one of the predominant indication for uterine removed in premenopausal women.Because of the high morbidity and poor curative effect of non-surgical treatment,as well as the populizing medical knowledge, more numberous women in clinical want to own the function of uterine and maintain their bodvs integrity.Therefore,there will be a great significance to develop the new non-surgical treatment of uterine fibroids.in the base of the clear cause and machnism of the disease. Though it is a disease,in which estrogen and progestogen have played an important role.no clear cause has found. Myocardin is now a key factor of the hypertrophy of myocardial cells.While there are evidences that NF-κB p65and Myocardin affected the growth、proliferation and differentiation of the smooth musule cells. In this study,we took uterine leiomyomas tissue and primary uterine leiomyoma cells as subjects, compared the difference of the expression of NF-κB p65and Myocardin,and investigated the correlation between them in the mechanism of uterine leiomyoma.Methods:At first, we used Western Blotting approach to examine the expression of NF-kB p65and Myocardin in uterine leiomyoma tissues followed by the correspondenting myometrium(n=29),as well as the primary uterine leiomyoma smooth muscle cells and the adjacent myometrium cells(n=15).Besides,we applied RNAi assay to depress the expression of NF-κB p65in the primary uterine leiomyoma smooth muscle cells.which observed by optical microscopy. Finally we examined the expression of NF-κB p65and Myocardin in those with Western Blotting approach.Statistical analysis was performed with the SPSS16.0software package.Results:1.Comparing with the normal myometrium.the expression of NF-κB p65in uterine leiomyoma tissues was higher(P<0.05),whereas Myocardin was lower(P<0.05),which is of statistical significance in the difference.2.Comparing with the normal myometrium.the expression of NF-κB p65in uterine leiomyoma tissues cells was higher(P<0.05),while Myocardin was lower(P<0.05), and the differences were statistically significant,similar to the tissue results.3. After RNA interference targeting for primary uterine leiomyoma cells,the expression of NF-κB p65was lower(.P<0.05),whereas Myocardin was higher(P<0.05),in contrast with the primary uterine leiomyoma smooth muscle cells without transfection.Conclusion:1.The high expression of NF-κB p65and the low expression of Myocardin in the tissue and cells of uterine leiomyoma suggested the both NF-κB and Myocardin may participate in the formation of uterine leiomyoma.2.The high expression of Myocardin and the low expression of NF-κB p65in the primary uterine leiomyoma cells with RNA interference suggested that NF-κB p65and Myocardin may interact with each other in the formation of uterine leiomayoma.
