ObjectiveTo identify the hepatic progenitor cell features of NCAM+c-Kit+subset cells in intrahepatic cholangiocarcinoma cell line RBE.MethodMagnetic activated cell sorting was used to isolate NCAM+c-Kit+/NCAM"c-Kit-RBE subset cells, which were tested for hepatic progenitor cell properties including the abilities of proliferation, differentiation, colony formation and initiate highly invasive in nude mice.Results1.The absorbance of NCAM+c-Kit+REB subset cells in490nm was stronger than that of NCAM-c-Kit-REB subset cells (P<0.01).2.In serum-free condition, the number of colonies formed by NCAM+c-Kit+subset cells was more than that of NCAM-c-Kit-subset cells(P<0.01).3.1×104NCAM+c-Kit+RBE cells were enough to form tumors in nude mice after subcutaneous inoculation for two weeks, while1×106NCAM-c-Kit-RBE cells were necessary to form tumors for three weeks. The tumor formation rate of NCAM+c-Kit+RBE cells was higher than that of NCAM-c-Kit-RBE cells(P=0.04).4.The rate of NCAM+c-Kit+RBE of the NCAM+c-Kit+tumor spheres which were cultivated in serum-free condition for15days was dramatic decline from (99.1%/98.5%) to (79.0%/55.3%).This suggests NCAM+c-Kit+RBE not only have the abilities of self-renew but also differentiate.Conclusion1. The cell proliferation ability of NCAM+c-Kit+RBE subset cells was stronger than that of NCAM-c-Kit-cells.2. The number of colonies formed by NCAM+c-Kit+RBE was more than NCAM-c-Kit-cells.3. The tumor formation rate of NCAM+c-Kit+RBE was higher than that of NCAM-c-Kit-RBE.4. NCAM+c-Kit+subset cells have the ability of differentiate.In a word, it is possible that NCAM+c-Kit+RBE subset cells have the properties of hepatic progenitor cells. NCAM combined with c-Kit might be a valuable marker for isolating and purifying ICC stem/progenitor cells.
