| Objective To investigate the expression of THBS2and its correlation withclinicopathologic features in gastric cancer.Methods The mRNA and protein expression levels of THBS2were assessed in14paired of gastric cancer specimen and corresponding normal mucosa collected from theEighth Department of General Surgery, The First Affiliated Hospital of Anhui MedicalUniversity using quantitative real-time PCR and Western blot analysis.129formalin-fixed, paraffin-embedded human gastric cancer and24randomly selectednormal gastric tissues consecutively recruited between December2006and May2008from Department of General Surgery, The First Affiliated Hospital of Anhui MedicalUniversity. Collected the information of them, and made these specimens into aTMA(Tissue microarray). Immunohistochemical staining was carried out for evaluatingTHBS2protein expression level. Then used SPSS15.0explore the association betweenclinicopathological variables and THBS2expression. The Kaplan–Meier method andCox’s proportional hazards model were used in survival analysis.Results Both mRNA and protein expression of THBS2in12gastric cancer tissues areremarkably lower than the corresponding normal tissues among total14pair tissues.Consistent with the results of our Western blot, THBS2expression was significantlyinhibited in gastric cancer tissues as that in the normal controls in TMA (P=0.031).TNM stage (P <0.001) and THBS2expression (P <0.009) were both independentprognostic factors for the assessment of patient outcomes. High TNM stage is a riskfactor for longer survival [P <0.001, HR=7.23,95%CI3.55-14.69). However,increasing THBS2expression is a favorable factor for longer survival (P=0.009, HR= 0.51,95%CI0.31-0.85). And the data also exhibited that decreasing THBS2expressionis associated with the poor histological grade of gastric cancer histological grade (P=0.005).Conclusion1) THBS2expression are remarkably lower in the gastric cancer tissue versus normalgastric tissue in both mRNA and protein level.2) Our data exhibited a significant negative correlation between THBS2expressionlevel and the histologic grade of gastric cancer.3) TNM stage and THBS2expression level were both independent prognostic factorsfor the assessment of patient outcomes. High TNM stage is a risk factor for longersurvival. However, increasing THBS2expression is a favorable factor for longersurvival. |
