Objective1. To observe the influence on the proliferation of T. asahii after inhibititing the lipidrafts and spitzenk rper formation.2. To observe the influence on pathogencity of T. asahii in murine model afterinhibititing the lipid rafts and spitzenk rper formation.Methods1. To inhibit the lipid rafts formation, the experimental group bacteria suspensionwere cultivated with different concentration of amphotericin B0.2u g/ml,0.5u g/ml,1.0ug/ml and2.0ug/ml respectively, relatively, the control group with the normal cellsuspension. In vitro, the growth curve and bacterial colony were measured. Meanwhile,each group was inoculated with T. asahii through the caudal vein of each mouse afterimmunosuppression with cyclophosphamide (200mg/kg of body weight/day for3days). Then, the mortality and infection rate of mice in3weeks were counted aftermain viscera of the mice were examined by means of tissue culture and pathologicsections.2. To inhibit the Spitzenk rper formation, the experimental group cell suspensionwere cultivated with different concentration of cytochalasin D0.5umol/ml,1.0umol/mland2.0umol/ml respectively, relatively, the control group with the normal cellsuspension. In vitro, the growth curve and bacterial colony were measured. Meanwhile,each group was inoculated with T. asahii through the caudal vein of each mouse afterimmunosuppression with cyclophosphamide (200mg/kg of body weight/day for3 days). Then, the mortality and infection rate of mice in3weeks were counted aftermain viscera of the mice were examined by means of tissue culture and pathologicsections.Results1. We confirmed the amphotericin B can inhibit the lipid rafts formation of T.asahii,and the treated cell suspention mainly exists in the form of spores and little hyphae.T.asahii polarity is affected by a certain growth.2. We confirmed the cytochalasin D can inhibit the Spitzenk rper formation of T.asahii,and the treated cell suspention mainly exists in the form of spores and little hyphae.T.asahii polarity is affected by a certain growth.3. After inhibitting the lipid rafts formation by amphotericin B, the formation of invitro culture cell growth was significantly delayed16h to24h, and the growingcolonies diameter decreased significantly too. Mainwhile, in vitro mortality in miceand the infection rate was decreased in different degree, with the increasing drugconcentration, its pathogenic decreased obviously.4. After inhibitting the Spitzenk rper formation by cytochalasin D, the formation of invitro culture cell growth was significantly delayed8h to16h, and the growing coloniesdiameter decreased significantly too. Mainwhile, in vitro mortality in mice and theinfection rate was decreased in different degree, with the increasing drug concentration,its pathogenic decreased obviously.Conclusions1. Amphotericin B and cytochalasin D can inhibit lipid rafts formation andSpitzenk rper formation of T.asahii respectively. This indicates that both of them canmake its polar growth abnormalities. And with the improving drug interventionconcentration, it even appears no growth phenomenon. 2. With the increasing of drugs concentration, T.asahii in vitro growth is restrainedmore obviously, as well as the lower the pathogenicity of mice. From this point, we cansuggest that lipid rafts and Spitzenk rper may be a clue for a new target of anti-fungaldrug that would be discovered.
