In China the denovo tumors assumes unceasingly the trend of escalationevery year according to the latest survey. The lung cancer, which incidenceranks first amongst malignant tumor, was the most common malignancy of therespiratory system and the death rate topped the list in the malignancy.Non-small cell lung cancer was the most common pathological type of lungcancer. Most patients died from lung cancer recurrence or distant metastasissoon or later, though the operation based, multidisciplinary and comprehensivetreatment of lung cancer got better results and the chemotherapeutic efficacywas significant increased by new chemotherapy drugs. Leading cause of death inpatients was the lung cancer invasion and metastasis.As the intense research on relative molecular mechanism of occurrence,development and invasion of lung cancer, many important substances werefound and proved which relative to development and invasion of lung cancer.Two of the most important substances were matrix metalproteinases(MMPs) andvascular endothelial growth factor(VEGF). MMPs,which accelerate invasionand metastasis of lung cancer by catabolited the proteins of the extracellular matrix (ECM), was a group of protease that highly dependent by Metal ion. Theimportant foundation of the formation and metastasis of lung cancer wasangiogenesis. VEGF played a significant role in development and invasion oflung cancer,which was a vital factor in regulation of blood vessel producing.MMPs and VEGF were confirmed closely linked to development,metastasis and prognosis of lung cancer thought massive domestic and foreignrelated research, but some of the mechanisms stillunknown.Immunohistochemistry and real-time PCR were used to detect theexpression of MMP-2,MMP-9and VEGF in human NSCLC tissues andparacarcinoma tissues,relatively few such studies. The aim of this research atresearching the expression of MMP-2, MMP-9and VEGF in non-small celllung carcinoma from the molecular level and the gene transcription level anddiscussing the relationship between the expression and the clinicopathologicalfeature.Objective: To study the expression of MMP2、MMP9and VEGF innon-small-cell lung carcinoma (NSCLC) and analyze the clinical significancebetween the expression of MMP-2,MMP-9and VEGF.Methods: Sixty-eight cases of non-small cell cancer tissues andparacarcinoma tissues,and50cases of the benign lesions tissues were collected,which excision from Thorscic Surgery of NO1.hosipital,Guangxi MedicalUniversity.The position rate of protein of MMP-2,MMP-9and VEGF weredetected by mmunohistochemistry,and the expression level of MMP-2mRNA,MMP-9mRNA and VEGFmRNA. Meanwhile, the clinical pathologicalinformation (include gender,age,smoking,lymphatic metastasis and distantmetastasis),then statistical analysis was performed.Results:1.The positive rate of protein in lung cancer tissues of MMP-2 was61.8%. The expression was related to NSCLC clinical stages(P=0.011),differentiation(P=0.025), lymphatic metastasis(P=0.017)and distantmetastasis(P=0.012). The expression level of MMP-2mRNA was consistentwith the expression result of positive rate of protein in NSCLC cancer tissue.The expression level of MMP-2in NSCLC cancer tissues were significantdifference compared with paracarcinoma tissues and the benign lesions tissues(P<0.05).2. The positive rate of protein in lung cancer tissues of MMP-9is58.3%.The expression is related to NSCLC clinical stages(P=0.001), differentiation(P=0.002), lymphatic metastasis(P=0.023)and distant metastasis(P=0.000). The expression level of MMP-9mRNA was consistent with theexpression result of positive rate of protein in NSCLC cancer tissue. Theexpression level of MMP-9in NSCLC cancer tissues were significant differencecompared with paracarcinoma tissues and the benign lesions tissues(P<0.05).3. The positive rate of protein in lung cancer tissues of VEGF is73.5%.The expression is related to NSCLC clinical stages(P=0.015), differentiation(P=0.000), lymphatic metastasis(P=0.014)and distant metastasis(P=0.016). The expression level of VEGF mRNA was consistent with theexpression result of positive rate of protein in NSCLC cancer tissue. Theexpression level of VEGF in NSCLC cancer tissues were significant differencecompared with paracarcinoma tissues and the benign lesions tissues(P<0.05).4. The expression of MMP-2、MMP-9、VEGF in NSCLC cancer tissuewere relevant.Conclutions:The results suggest that MMP-2,MMP-9and VEGF paly veryimportant roles in carcinogenesis of NSCLC.The over expression of theMMP-2,MMP-9and VEGF gene were correlated with invasion and metastasis of NSCLC.The expression of MMP-2,MMP-9and VEGF could worktogether for cancer invasion and metastasis....
