Expression Of T Helper17(th17) Cells, Th22Cells And Interleukin-22(IL-22) In Peripheral Plasma And Deciduas Of Unexplained Recurrent Spontaneous Abortion

Objective:Through the detectionof the proportions of T helper17(Th17)cells and Th22cells and interleukin-22(IL-22) levelsin peripheral plasma anddeciduas of unexplained recurrent spontaneous abortion(URSA), to understandthe changes of the proportions of Th17cells and Th22cells and IL-22levelsinperipheral plasma and deciduas ofURSA.Method：A total of30patients participated in this study,15patients in theURSA group and15normal pregnancy(NP) women who required artificialabortion operation in the control group. We drawn the elbow blood5ml beforethe abortion operations, and collected the decidua tissues after the operations.We detected the proportions of Th17cells and Th22cells and IL-22levelsinperipheral plasma and deciduas by flow cytometer and enzyme-linkedimmunosorbent assay(ELISA).Results:1、The proportions of Th17cells in peripheral blood and deciduatissue in the URSA group(1.62%±0.52%and2.11%±0.61%) were significantly than that in the NP group(1.24%±0.27%and1.47%±0.41%)（P<0.05）. AndTh17cells proportions in decidua tissue were higher than that in peripheralblood, the difference was statistically significant（P<0.05）.2、The proportions of Th22cells in peripheral blood and decidua tissue inthe URSA group(1.59%±0.52%and1.91%±0.58%) were significantly than thatin the NP group(1.11%±0.24%and1.33%±0.32%)（P<0.01）. And Th22cellsproportions in deciduas tissue were higher than that in peipheral blood, thedifference was statistically significant（P<0.05）.3、The IL-22levels in peripheral blood and deciduas tissue in the URSAgroup(181.98±8.10pg/ml and198.52±10.98pg/ml) were significantly than thatin the NP group(104.28±4.73pg/ml and110.47±6.43pg/ml)（P<0.01）. AndIL-22levels in deciduas tissue were higher than that in peripheral blood, thedifference was statistically significant（P<0.01）.4、Compared the correlations of IL-22levels and Th17proportions in theURSA group and the NP group, the correlation coefficient and P values were: inthe peripheral blood(r=0.157, P<0.05),(r=0.437, P>0.05); in the deciduas(r=0.069, P<0.05),(r=0.235, P>0.05). There were extremely weak or nocorrelations statistically.5、Compared the correlations of IL-22levels and Th22proportions in theURSA group and the NP group, the correlation coefficient and P values were: inthe peripheral blood(r=0.885,P<0.05),(r=0.749, P<0.01); in the deciduas(r=0.849, P<0.05),(r=0.934, P<0.01). There were positively correlatedstatistically.6、In the URSA group and the NP group, compared the correlations ofTh17proportions in blood and deciduas, the correlation coefficient and P values were:(r=0.661, P<0.01),(r=0.954, P<0.01). There were positively correlatedstatistically.7、In the URSA group and the NP group, compared the correlations ofTh22proportions in blood and deciduas, the correlation coefficient and P valueswere:(r=0.685, P<0.01),(r=0.943, P<0.01). There were positively correlatedstatistically.8、In the URSA group and the NP group, compared the correlations ofIL-22levels in blood and deciduas, the correlation coefficient and P values were:(r=0.875, P<0.01),(r=0.823, P<0.01). There were positively correlatedstatistically.Conclusions:1、The high proportions of Th17and Th22cells and IL-22levels in blood and deciduas may associated with the occurrence of URSA. Andcompared with that in blood,those changes in deciduas may have closerrelationship with the pregnancy status.2、The proportions of Th17and Th22cells and IL-22levelsin blood and indeciduas were in parallel.3、The Th17and Th22cells proportions and IL-22levels in blood anddeciduas during pregnancy may be a predictor of the occurrence of URSA.
