Objective:To investigated the effects of Src family kinases on the expression of mRNA and protein of Nav1.1in spiral ganglion neurons, and explored the role of Src family kinases in the process of the regulation of voltage-gated sodium channels in spiral ganglion neurons, as to provide the theoretical basis in the audiology researchMethods:RT-PCR and Western blot techniques respectively explored the level of expression of mRNA and protein of Nav1.1in spiral ganglion neurons by Src inhibitorResults:A application of the inhibitor of Src family kinases which was PP2(10μM) and SU6656(2μM) gived rise to the mRNA decrease to (26±0.8)%and (36±1.5)%respectively (P<0.05), protein reduced to (39±12.5)%and (53±1.7)%severally(P<0.05), but under the action of the PP3, the change of the mRNA and protein both were not obvious, which were (80±5.4)%and (83±19.1)%respectively (P>0.05).Conclusion:Inhibition of Src family kinases could decrease the expression level of mRNA and protein of Nav1.1in spiral ganglion neurons. In other word, the inhibitor of Src family kinases which was PP2and SU6656mediated Nav1.1 by the Src family, and had nothing to do with their chemical structure itself That means that the Src family kinases directly participated in the regulation of Nav1.1on the auditory nerve system.
