Objective:This experiment aims to research follistatin-like protein1(FSTL1) stimulate the mature dendritic cells (DC) of patients withnasopharyngeal carcinoma. And then investigate the impact of the DC’s immunefunction on FSTL1. Explore the potential about FSTL1as cancerimmunotherapy drugs.Methods: Collected peripheral blood25ml from17nasopharyngealcarcinoma patients and10healthy volunteers, and isolated mononuclear cells(PBMC). Separated monocytes and lymphocytes by adherent. There wereHLA-A2+monocytes of8NPC patients and6healthy volunteers screened byflow then continue cultured. Lymphocytes were cryopreserved if used insometimes. Monocytes were induced to mature DC by rhGM-CSF (1000U/ml),rhIL-4(500U/ml) and rhTNF-α (1000U/ml), and phenotype include CD40, CD86and HLA-DR were detected in the first three days and eight days.Collected mature DC and divided into two groups: the first group received notreatment, scilicet DC group; the second group joined FSTL1as stimulus,scilicet DC+FSTL1group. Two groups of DC presented antigenic peptides FLY,and then DC and autologous lymphocyte by recovery were blended cultured.Cells were collected in the first fifteen days. Detected DC antigen-presentingand stimulated lymphocytes turn into specific lymphocytes by Elispot andTetramer.Results:1.We successfully induced maturation of dendritic cells, irregular shapeand many different lengths of the projections, the form was dendritic.2.Elispot results:①Nasopharyngeal carcinoma patients group: The specific lymphocytecounts (scilicet number of spots) which secreted IFN-γ of DC group and DC+FSTL1group medians were40.00(11.00-93.00),51.00(31.00-118.00). Thedifferences were statistically significant (P <0.05);②normal control group:The specific lymphocyte counts (scilicet number of spots) which secreted IFN-γof DC group and DC+FSTL1group medians were59.50(34.33-88.67),72.00(41.00-104.00). The differences were statistically significant (P <0.05);③DC+FSTL1group between NPC patients and normal control were compared andthe differences were not statistically significant (P>0.05). 3.Tetramer results:①Nasopharyngeal carcinoma patients group: The specific lymphocytecounts(%) of DC group and DC+FSTL1group medians were0.86(0.15-2.09),1.38(0.41-3.82). The differences were statistically significant (P<0.05);②normal control groups: The specific lymphocyte counts(%) of DC group and DC+FSTL1group medians were1.75(0.13-2.60),2.63(0.23-3.75). Thedifferences were statistically significant (P<0.05);③DC+FSTL1groupbetween NPC patients and normal control were compared and the differenceswere not statistically significant (P>0.05).Conclusion:FSTL1could improve immune function of dendritic cells innasopharyngeal carcinoma patients.
