Identification Of Ncrna Profile And Screen Of Mirna In Semen Of ⅢA CP/CPPs Patients

Objective:To detect the ncRNA profile in ⅢA CP/CPPS patients’semen, miRNAs were screened in semen samples from ⅢA CP/CPPS patients and healthy normal adults respectively.Methods:The semen samples from ⅢA CP/CPPS patients were screened as disease group, and the semen from healthy adults semen were collected as control group, total RNAs were extracted respectively. ncRNA were collected through the recycle of glue,3’and5’adaptor connector were introduced, ncRNA cDNA library was established by RT-PCR method, high-throughput sequence technology of solexa was adopted to analysis the ncRNA expression in semen from two groups respectively, miRNAs differentially expressed in sperm with normal semen were analysed comparatively, the main differences expression of miRNAs molecules were selected.Results:1.1341489sequences were found from the comparation beween unique sequences in disease group and the ncRNA in Genbank, rRNA117574, scRNA7300, snRNA4347,4347, snoRNA889, tRNA49431, other sequences1161948;39833420sequences were found from the comparation beween total sequences in disease group and the ncRNA in Genbank, rRNA13484159, scRNA1017352, snRNA328085, snoRNA5364, tRNA5245222, other sequences19753238.2. Variance analysis was conducted between disease group and control group. The expression of52miRNAs were up-regulated,156down-regulated, the remaining278miRNAs showed no apparent changes according to variance analysis. There were molecules expressed differenent apparently, such as the miR-203, the miR-363, the miR-363, the miR-664, the miR-146, the miR-25, the miR-423. Some molecules showed up-regulated expression, such as miR-378, the miR-203, the miR-185, the miR-181, other molecules showed down-regulated expression, the miR-24, the miR-363, the miR-221, etc.Conclusion:1. In this study, we found that miRNA had higher concentration than other ncRNA molecules in semens of ⅢA CP/CPPS patients, and miRNA can be used as marker for the research of ⅢA CP/CPPS patients.2. Acorrding to the screen of486miRNAs in semen from IIIA CP/CPPS patients, we found that the expression of52miRNA up-regulated, and156down-regulated, the remaining278miRNAs showed no apparent changes by variance analysis.3. The miRNA selected may be related to the occurrence and development of ⅢA CP/CPPS.
