Object:To explore the regulation effect of1,25(OH)2D3on IL-4in rat with IgA Nephropathy.Methods:(1)42SD rats, BSA, CCL4and LPS were used to establish IgA nephropathy model. Normal control group was intragastric administration and subcutaneous injection and tail vein injection with the same dose of NS.(2)IgA nephropathy model was divided into4groups:Model group(Agroup),prednisone treatment group, prednisone+l,25(OH)2D3treatment group,1,25(OH)2D3treatment group.1,25(OH)2D3was intragastric administration in3ng/100g on morning per day; prednisone was intragastric administration in10mg/kg on morning per day. Model group and normal control group intragastric administration with the same dose of NS. After two weeks urine protein of rats in24h, urine red blood cells and change of renal IgA deposition were detected. The spleen and lymph nodes Th2cell level was detected with flow cytometry.Result:(1) The24h urine protein of model group were51.49±3.04mg/24h; the24h urine protein of prednisone treatment group were12.15±0.75mg/24h; the24h urine protein of prednisone+1,25(OH)2D3treatment group was9.99±0.79mg/24h; the24h urine protein of1,25(OH)2D3treatment group were23.0±2.27mg/24h; the24h urine protein of normal control group were7.03±0.99mg/24h. The24h urine protein of model group was higher than prednisone treatment group, prednisone+1,25(OH)2D3treatment group,1,25(OH)2D3treatment group and control group(p<0.05). After treatment the24h urine protein of each group were declined significantly (p<0.05). The prednisone+1,25(OH)2D3treatment group declined significantly compare to prednisone treatment group,1,25(OH)2D3treatment group(p<0.05).(2) IL-4level on spleen of model group were5.10±0.08%; IL-4level of prednisone treatment group were2.93±0.13%; IL-4level on spleen of prednisone+1,25(OH)2D3treatment group were1.44±0.56%; IL-4level on spleen of1,25(OH)2D3treatment group were4.56±0.19%; IL-4level on spleen of normal control group were1.43±0.60%; IL-4level on spleen of model group were higher than normal control group(p<0.05). After treatment IL-4level of each group declined significantly. The prednisone+1,25(OH)2D3treatment group declined significantly compare to prednisone treatment group,1,25(OH)2D3treatment group, model group(p>0.05), but there is no significant difference between prednisone+1l,25(OH)2D3treatment group and normal control group(p>0.05).(3) IL-4level on lymph nodes of model group were15.4±3.11%; IL-4level on lymph nodes of prednisone treatment group were2.74±0.29%; IL-4level on lymph nodes of prednisone+1,25(OH)2D3treatment group were2.32±0.34%; IL-4level on lymph nodes of1,25(OH)2D3treatment group were6.55±0.66%; IL-4level on lymph nodes of control group were1.50±0.13%. After treatment the IL-4level on lymph nodes of each group declined respectively. The prednisone+1,25(OH)2D3treatment group declined significantly compare to prednisone treatment group,1,25(OH)2D3treatment group, model group(p>0.05), but there is no significant difference between prednisone+1,25(OH)2D3treatment group and normal control group(p>0.05).Conclusions:IL-4level in IgAN might elevated,1,25(OH)2D3might downregulated the expression of IL-4in IgAN and then affect Th2cell regulation.
