Objective:Compare the differences of microRNA expression profile in multiple glioma and solitary glioma tissues which were formalin-fixed and paraffin embedded (FFPE), explore the role of difference expressed microRNA and their target genes in invasion and migration progress.Method:Paraffin embedded multiple glioma(n=6) and solitary glioma(n=6) tissue sanples were collected for this study, execute routine section and extract total RNA for quanlity test. Detect microRNA expression profile by using human microRNA testing chip producted by Agilent Company, and analyse the datas by using gene analysis software in order to obtain difference expressed mircoRNA in the two kinds of tissues, then we predict the target genes of difference expressed mircoRNA through bioinformatics software.Result:All the RNA extraced from formalin-fixed and paraffin embedded tissues were reach the standard of quanlity control. We discovered that the expression of128kinds of microRNA obviously changed in multiple glioma tissues compared with solitary glioma (multiple greater than5or less than0.5), and ten of them significantly upregulatedmiR-21,miR-455,miR-199.miR-410,miR-148a,miR-200b,miR-34b,miR-664a,miR-542,miR-4453. Meanwhile, another5of them downregulated (multiple less than0.05):miR-146, miR-451, miR-29a, miR-101, miR-96. The predicted target genes based on part of the difference expressed microRNA mainly conserned the functions such as nucleic acid connection, protein connection, transferase activity and epithele inversion, which played roles in tumor invasion and migration proccesses.Conclusion:MicroRNA difference expressed in multiple glioma tissues compared with solitary glioma, and miR-21/miR-200a might closely related to tumor invasion and migration proccesses, and it is hopeful to find new target genes for tumor diagnosis and therapy in furter study incuding a large sample. Formalin-fixed and paraffin-embedded tissue might be an abundant resource for bioinformatics function studies or exploration.
