Objective:To investigate the protection of diabetic nephropathy in rats on Tripterygium wifordiipolyglucosides and its effect on Wnt/β-catenin signaling pathway.Methords:40healthy male Wistar rats were randomly divided into NC group (n=10), DN group (n=10),DT group (n=10)and DI group (n=10). The animal model of Diabetic nephropathy wereestablished by injection of streptozotocin in abdominal.After the modeling was completed,DTgroup and DI group were administered by intragastric injection with Tripterygium wifordiipolyglucosides(0.5mg/100g.d) and Irbesartan(17.5mg/Kg.d). NC group and DN group wereadministered by the same amount of citrate buffer solution.The24hPro quantitative weredetected respectively at the end of4w、8w、12w、16w and record24hour urine volume.At theend of16weekend, Glu、Scr and BUN were detected and record kidney weight and body weight.HE and PAS staining was performed on renal tissue to observe kidney tissue pathology;theexpression of wnt4, β-catenin and IV collagen in renal interstitium were detected byimmunohistochemistry; and the expression level of wnt4, β-catenin and IV collagen weredetected via real-time fluorescence quantification PCR method.Results:1. At the end of16thweek, Glu, Scr and BUN of the rats in DN group, DT group and DI groupshowed an increase compared with that of NC group, with statistical significance of differences(P<0.05);compared with DN group, the differences of all indicators of DT group and DI groupindicated no statistical significance (P>0.05).2.24h urine protein quantitation of DN group showed an increased compared with NC group(P<0.05); compared with DN group,24h urine protein quantitation of DT and DI groupsdeclined obviously (P<0.05), but the differences of DT and DI groups showed no statisticalsignificance (P>0.05).3. kidney weight/body weight (KW/BW) of DN group showed an increased compared withNC group (P<0.05); compared with DN group, KW/BW of DT and DI groups declinedobviously (P<0.05), but the differences of DT and DI groups showed no statistical significance(P>0.05).4. Light microscope: no obvious abnormality was discovered in NC group; however in DNgroup, Part of the renal tubular epithelial cell degeneration, loss, tubulointerstitial area increased stromal matrix between the endoplasmic see a lot of inflammatory cell infiltration, renalinterstitial fibrosis visible. And after Tripterygium glycosides and irbesartan treatment, renaltubular epithelial cell degeneration seen the situation improve, tubulointerstitial stromalhyperplasia eased, interstitial infiltration area to reduce inflammatory cells, fibrosis obvious.5. Immunohistochemical result: compared with DN group, the expression of wnt4, β-cateninand collagen IV in renal interstitium of DT and DI groups rose obviously (P<0.05), with nostatistical significance of the differences between DT group and DI group (P>0.05). Correlationindicates: DN rats collagen IV expression was positively correlated with Wnt4(r=0.705, P<0.05), the expression of collagen IV and β-catenin was positively correlated (r=0.713, P <0.05).6. Real-time fluorescence quantification PCR display; compared with DN group, theexpression of wnt4, β-catenin and collagen IV mRNA in renal interstitium of DT and DI groupsrose obviously (P<0.05), with no statistical significance of the differences between DT groupand DI group (P>0.05).Conclusion:1. Tripterygium wifordii polyglucosides can lower the24h urine protein level of DN rats,restrain renal hypertrophy, relieve pathological damage on kidney.2. Ttripterygium wifordii polyglucosides may inhibit the Wnt/β-catenin signaling pathway,reducing the expression of collagen IV, and delay the progression of renal interstitial fibrosis inrats with DN, and thus play a role in protecting the kidneys.
